Identification of Bacillus Strains Using the API System

Abstract: A system is described for the rapid and accurate identification of Bacillus isolates using a matrix of results from tests in the API 20E and API 50CHB strips and from supplementary tests. API System tests have been shown to be more reproducible than the classical tests. A taxonomy based upon API tests is in good agreement with those obtained by other methods. The results matrix can also be used in computer assisted identification.

“…Isolate Dac521, a Gram-positive, sporulating, nonmotile, rod-shaped organism (2.0 x 0.7 pm) which grew between 35 and 70 "C on solid media and up to 73 "C in liquid media, was identified as a thermophilic Bacillus species using the API 5OCH and 20E identification test strips (Logan & Berkeley, 1984). On nutrient agar plates, Dac521 colonies were of non-uniform size, circular form, convex elevation, with an entire margin and a yellow/grey colour.…”

“…The API 20E, API 20NE, API Staph and API 50CH were designed for identifying members of the Enterobacteriaceae, Nonenteric Gram-negative rods; Staphylococcus and Bacillus respectively. APIWEB software was used for identification and was considered acceptable when given a probability of 80% or greater (BioMerieux, France) (Awong-Taylor et al, 2008;Logan and Berkeley, 1984).…”
