Identification of Proteins and Peptide Biomarkers for Detecting Banned Processed Animal Proteins (PAPs) in Meat and Bone Meal by Mass Spectrometry

Abstract: The outbreak of bovine spongiform encephalopathy (BSE) in the United Kingdom in 1986, with processed animal proteins (PAPs) as the main vector of the disease, has led to their prohibition in feed. The progressive release of the feed ban required the development of new analytical methods to determine the exact origin of PAPs from meat and bone meal. We set up a promising MS-based method to determine the species and the source (legal or not) present in PAPs: a TCA-acetone protein extraction followed by a cleanup… Show more

“…Tricine-SDS-PAGE gels (4% spacer gel and 16% separating gel) were used to resolve the peptide extracts. Proteins were separated by SDS-PAGE (5% spacer gel and 10% separating gel) [ 26 , 27 ]. The results were analyzed by Quantity One version 4.6.2 (Bio-Rad Technical Service Department, Oakland, CA, USA).…”

“…One notable issue relates to the erroneous detection of pig blood products (RIKILT, unpublished results). It appeared to be difficult to detect bovine specific markers based on peptide sequences, detectable at 1%, in porcine and vegetal matrices (Marbaix et al 2016). In the framework of a project with publicprivate cooperation RIKILT tried to develop a poultry specific antibody with no success until now.…”

“…Premixed Protein Standard (44.3–200 kDa) of Takara Co. Ltd. (Dalian, China) was used for finding the relative molecular weight. The samples were mixed with 10 µL of electrophoretic loading buffer and heated for 5 min in a boiling water bath [ 26 , 27 ]. Different samples were separated by centrifugation at 8000 rpm for 15 min.…”
