Identification of transient microglial cell colonies in the forebrain white matter of developing rats

Abstract: Herein, we describe the existence of distinct colonies of transient microglial cells that reside in well-defined zones of the forebrain white matter. Rats, aged at postnatal day (P) 0, P2, P5, P7, P10, P15 or adult, were anaesthetised with halothane gas, and various neural centres were injected unilaterally with the tracer biotinylated Dextran. The neural centres injected were cingulate or sensorimotor cortices, ventral nuclei of the dorsal thalamus, and the pontine reticular formation of the brainstem. Rats w… Show more

“…Further, a single injection of tracer labels these amoeboid cells across the full thickness of the internal capsule (Earle and Mitrofanis 1997; this study). The perireticular amoeboid cells start to disappear from about 2 weeks after birth until none are left in the adult (Earle and Mitrofanis 1997).…”

“…We used DiI (in vitro), latex beads and CTB since they -unlike HRP, WGA-HRP, dextran or other substances (Leong et al 1983;Xu and Ling 1994a, b;Earle and Mitrofanis 1997) -were not readily accumulated by the large group of amoeboid microglial cells that resides in the perireticular nucleus during postnatal development. We also made several injections of biotinylated dextran ( Fig.…”

“…Findings in kittens suggest that the presence of (Innocenti et al 1983a). However, the exact significance of these clusters remains unproven, as microglial cells have been reported to accumulate at specific white-matter sites after the corresponding axons reach their final target (Earle & Mitrofanis, 1997).…”
