IFN-γ Drives Human Monocyte Differentiation into Highly Proinflammatory Macrophages That Resemble a Phenotype Relevant to Psoriasis

Abstract: As key cells of the immune system, macrophages coordinate the activation and regulation of the immune response. Macrophages present a complex phenotype that can vary from homeostatic, proinflammatory, and profibrotic to anti-inflammatory phenotypes. The factors that drive the differentiation from monocyte to macrophage largely define the resultant phenotype, as has been shown by the differences found in M-CSF– and GM-CSF–derived macrophages. We explored alternative inflammatory mediators that could be used for… Show more

“…Macrophages are highly plastic immune cells that exhibit pleiotropic and coordinated responses to various microenvironmental signals [1][2][3]. They present a complex phenotype that can vary from homeostatic, pro-inflammatory, and profibrotic to reparative phenotypes [4]. During the natural wound-healing process, macrophages initially exhibit a pro-inflammatory phenotype, which is vital for killing microbes but leads to widespread damage to the surrounding tissues [5,6].…”

“…Next, we performed automated high content image-based fluorescence screening in non-activated and phagocytic bone marrow-derived macrophages (BMDMs) to identify NIR fluorophores that would preferentially emit in phagocytic cells with enhanced trafficking activity. We ran a fluorescence-based screening where we cultured BMDMs in the absence or presence of interferon (IFN)-γ, 57 followed by incubation with the NIRBD dyes. We acquired fluorescence microscopy images, using the ImageXpress high content screening platform, in both non-activated and IFNγ-activated cells and identified NIRBD dyes (NIRBD-22,26,48,62) with brighter emission in phagocytic cells ( Fig.…”

“…PSO pathogenesis has not been fully elucidated[ 16 ]. Excessive activation of the adaptive immune system is considered to be at the core of PSO[ 17 ]. Early during PSO, various cell types secrete cytokines to activate myeloid dendritic cells.…”
