IgE‐induced histamine release from rat basophilic leukemia cell lines: isolation of releasing and nonreleasing clones

Abstract: The rat basophilic leukemia (RBL) cell lines were cloned and the various sublines compared for their chromosome number, IgE-mediated histamine release and for IgE surface receptors. It was found that cell lines started from tumors at different times vary in both their chromosome number and their ability to release histamine by an IgE-mediated reaction. RBL-I and III have approximately 44 chromosomes and did not respond to an IgE-mediated reaction. RBL-II and RBL-IV have 68-73 chromosomes and showed moderate le… Show more

“…RBL2H3 cells retain this ability to respond [14,15]; the cells discharged about 20% of the cellular serotonin (total: = 1.75 nmol/106 cells) within 1 h. A calcium ionophore A23187 (0.08 ~tM) caused serotonin release by RBL cells similar to that described for histamine release [10]. After immune stimulation (DNP-BSA/IgE), the TPH activity of RBL2H3 cells increased up to about 9-fold (Fig.…”

“…We have recently shown that direct nerve-mast cell (RBL) cross-talk can occur in the absence of an intermediary transducing cell and that the neuropeptide substance P is an important mediator of this communication. 6,17) It is known that RBL cells have the properties of MMCs and do not respond to substance P. 18,19) We tried to stimulate wild-type RBL cells with substance P (0.1-10 mM), but they did not respond. However, the addition of purified neutralizing anti-substance P to the co-culture milieu of the neurite-RBL system dose dependently prevented RBL activation elicited by bradykinin or scorpion venom stimulation of neurites, but did not have any direct effect on neuronal activation.…”

“…The E5 cells express 82.9% and D1 cells express <1% of GM1 in relation to the amount in RBL-2H3 cells (Silveira e Souza et al 2010). All cell lines used in these experiments were grown as monolayers, as previously described (Barsumian et al 1981).…”
