IGF-I Transcript Levels in Whole-Liver Tissue, in Freshly Isolated Hepatocytes, and in Cultured Hepatocytes from Lean and Obese Zucker Rats

Abstract: Background: The mechanisms underlying the maintenance of normal to high rates of linear growth and plasma insulin-like growth factor I (IGF-I) levels in spite of a low growth hormone secretion in obese children remain unknown. Among the animal models of early-onset obesity, obese Zucker (fa/fa) rats (which are homozygous for an inactivating missense mutation in the leptin receptor) are particularly appropriate, because their linear growth shows this growth hormone independence. Methods: To study the regulation… Show more

“…IGF‐1 gene transcription, in particular, is upregulated by CREB in osteoblasts and mesenchymal cells . Under normal and pathological conditions (e.g. tumor development and progression of a variety of different cancers such as pancreatic carcinoma, colon carcinoma, esophageal carcinoma and non‐small cell lung cancer), hepatocytes produce a variety of agents, including IGF‐1 and HGF.…”

“…(11) Briefly, the livers were perfused with 200 ml of Hank’s buffer (pH 7.5), followed by the same volume of collagenase buffer (pH 7.5). The livers were then isolated and placed in DMEM-F12 (−) FBS medium (Sigma-Aldrich Co.…”

“…The isolated hepatocytes were seeded at a density of 1.0 × 10 5 cells per 60 mm dish (coated with collagen-I) in DMEM-F12 medium supplemented with T 3 (final concentration: 1 × 10 −9  M), hydrocortisone (final concentration: 5 × 10 −8  M), ethanolamine (final concentration: 1 × 10 −6  M), ornithine (final concentration: 4 × 10 −4  M), lactic acid (final concentration: 1.8 × 10 −5  M), and penicillin/streptomycin (final concentration: 100 U/ml, 100 µg/ml). (10) After cell attachment (4 h), the medium was changed and the hepatocytes were incubated at 37°C in a 5% CO 2 atmosphere.…”
