IL-17 receptor signaling influences virus-induced corneal inflammation

Abstract: IL-17 has been associated with selected inflammatory and autoimmune diseases. We characterized the expression of this proinflammatory cytokine following HSV-1 corneal infection and investigated whether IL-17R signaling modulated the host response to the viral pathogen at early time-points postinfection. IL-17 was elevated in the murine cornea 24 h after high-dose virus infection and subsequently persisted at low levels during the first week. Immunofluorescent studies showed that the IL-17R was expressed by cul… Show more

“…IL-17 has been shown to be one of the major cytokines involved in the recruitment of neutrophils in several experimental models of inflammation (35,46,47). Our results show that in IL-17R gene-deficient mice marked diminution of neutrophil migration to the infection focus during NS-CLP-induced sepsis and, as a result, had a significantly lower survival rate.…”

“…IL-17 has been detected in the cornea of mice infected with HSV-1 and mice lacking IL-17R had a transient decrease in neutrophil infiltration, but corneal pathology and viral clearance were not significantly different than in wild-type mice [95]. We have recently found that antigenspecific Th17 cells are induced during infection with hepatitis C virus and that these are regulated by innate IL-10 and TGF-b induced in monocytes by the NS4 protein of the virus [34].…”

“…21,22 Th17 cells are the newly recognized CD4 þ T-cell subset, which mainly produces cytokines, including IL-17A, IL-17F, and IL-22, and is involved in a wide range of autoimmune diseases. [11][12][13] The level of IL-17 was found to be correlated with both activity and severity of various autoimmune diseases. Studies conducted on experimental mice model of DE have found that the level of IL-17 was significantly increased on the ocular surface and in tears.…”

“…Both IL-17 and IL-22 are the effective cytokines of Th17 cells, the levels of which, in tears, may represent the immune response of Th17 cells during the pathogenesis of DED. [11][12][13] In this study, we evaluated the levels of IL-17 and IL-22 in tears of patients with Sjögren's or non-Sjögren's syndrome DED. In addition, the correlation between disease severity and IL-17 as well as IL-22 levels in patients was also analyzed.…”
