IL-18 Contributes to Host Resistance Against Infection withCryptococcus neoformansin Mice with Defective IL-12 Synthesis Through Induction of IFN-γ Production by NK Cells

Abstract: The aim of this study was to examine the contribution of IL-18 in host defense against infection caused by Cryptococcus neoformans in mice with defective IL-12 production. Experiments were conducted in mice with a targeted disruption of the gene for IL-12p40 subunit (IL-12p40−/− mice). In these mice, host resistance was impaired, as shown by increased number of organisms in both lungs and brains, compared with control mice. Serum IFN-γ was still detected in these mice at a considerable level (20–30% of that in… Show more

“…7,11 This pulmonary cryptococcosis model is a long-term model compared with other published murine models. 8,[23][24][25][26][27] To monitor IL-4 production over time in this long-term model, IL-4 reporter mice, termed 4get mice, were infected, and enhanced green fluorescent protein (eGFP) expression, which is known to correlate with IL-4 transcription, was assessed. 20 There were constitutively eGFP ϩ pulmonary leukocytes in naïve mice ( Figures 1A and 2, A and B), as published by others.…”

“…Important insight into the role of NK cells in immunity to malaria was provided on IL-12-related mechanisms in early stages of the disease in the model. 42 Indeed, infection of mice can be ameliorated by administration of IL-12 early in the course of the infection. Untreated mice died 12 days after infection, while IL-12 treated mice survived.…”

“…Our results with p35,IL-4 Ϫ/Ϫ mice underscore the central role IL-12 plays in resistance to L. major. Resistance to several bacterial and viral infections can be IL-12-independent, although IFN-␥-dependent (45)(46)(47)(48)(49)(50)(51). It was conceivable that in the absence of an IL-4-mediated Th2 response, sufficient IFN-␥ might be produced by IL-12-independent mechanisms to promote resistance.…”
