IL-27 Activates Human Monocytes via STAT1 and Suppresses IL-10 Production but the Inflammatory Functions of IL-27 Are Abrogated by TLRs and p38

Abstract: IL-27 is a member of the IL-12 family of cytokines that activates the Jak-STAT signaling pathway in a context-dependent manner and has pleiotropic effects on acquired immunity. IL-27 has the capacity to promote early stages of Th1 generation, but recent evidence has suggested a predominant suppressive effect on Th1, Th2, and Th17 differentiation. Although modest suppressive effects of IL-27 on myeloid lineage cells have been observed, there is limited knowledge about the role of IL-27 in the regulation of inna… Show more

“…Expression of SOCS3 is slightly modulated by IL-27. Induction of SOCS expression in human T cells by IL-27 is a salient finding and is in concert with activation of STAT1 and SOCS1 in human macrophages (Kalliolias and Ivashkiv 2008). Whether the IL-27-mediated IL-17 inhibition is attributed to a SOCS1-mediated dampening of STAT1 or STAT3 and the role of SOCS3 remains to be determined.…”

“…43,59 However, this effect might depend on the target cells and on the microenvironment. 66 IL-27 polymorphisms affecting IL-27 have been associated to an increased susceptibility to ovarian cancer. 67 As proposed by Kalliolas et al, 66 IL-27 may exhibit opposite effects on innate and adaptive immune cells.…”

It is more likely that monocytes/ macrophages express IL-27 upon stimulation.…”

“…Although not responsible for the differentiation of alternatively activated macrophages, IL-27 may play an important role in the modulation of alternatively activated macrophage function (34). In contrast, IL-27 augments production of TNF-a, IL-12, IL-6, and IFN-g by human monocytes/macrophages (35,36).…”
