IL17 factors are early regulators in the gut epithelium during inflammatory response to Vibrio in the sea urchin larva

Abstract: IL17 cytokines are central mediators of mammalian immunity. In vertebrates, these factors derive from diverse cellular sources. Sea urchins share a molecular heritage with chordates that includes the IL17 system. Here, we characterize the role of epithelial expression of IL17 in the larval gut-associated immune response. The purple sea urchin genome encodes 10 IL17 subfamilies (35 genes) and 2 IL17 receptors. Most of these subfamilies are conserved throughout echinoderms. Two IL17 subfamilies are sequentially … Show more

“…The IL-17 family plays a critical role in the immune response in infectious, inflammatory, autoimmune, and neoplastic disorders [81,82]. The IL-17 family and its receptors are evolutionally ancient, and they are present in species as early as lamprey and sea urchins [83,84]. IL-17 family members comprise IL-17A, IL-17B, IL-17C, IL-17D, IL-17E, IL-17F, and IL-17AF [81,82].…”

“…However, the genome sequence contains homologs of tumor necrosis factor α, macrophage inhibitory factor and interleukin 17 (IL17), as well as IL1 receptors ( 23 ). This shared regulatory heritage between echinoderms and vertebrates enables experimental investigations into transcriptional control of immune cell development ( 25 , 26 ) and immune response ( 17 , 29 ) that can provide meaningful insight to vertebrate biology.…”

“…To identify an early immune signal, an RNA-Seq assay was used to quantify system-wide transcript levels in larvae over a time-course of exposure to V. diazotrophicus . From these data, a small family of genes orthologous to vertebrate IL17 cytokines emerged as the most highly upregulated transcripts across the entire genome ( 29 ). The mammalian IL17 signaling molecules [IL17A–F ( 48 )] are expressed in Th17 cells, and other lymphocytes, myeloid cell types, and barrier tissues ( 49 51 ), including gut epithelial cells ( 52 54 ).…”

“…The role that the sea urchin microbiome plays in immunity has received less attention [17,18]. In other invertebrate taxa, such as insects, the microbiome plays a role in protecting against pathogenic bacteria, while at the same time hosting commensal bacteria [19].…”
