Immune cells and bone: coupling goes both ways

Abstract: There is a mounting body of research describing how cells of the immune system direct bone cell function whilst bone cells are involved in the generation and retention of immune cells and their precursors. Recent works into regulation of the haemopoietic stem cell niche have firmly implicated osteoblasts and osteoclasts. On the other hand, virtually all of the mature immune cells have been described to influence bone formation in vitro and in vivo. This review will summarize the latest developments and discuss… Show more

“…In addition to osteoblasts and osteocytes, activated T cells, B cells, NK cells, fibroblasts, and thymus cells can also produce RANKL. Apart from osteoclasts and their progenitor cells, dendritic cells also express RANK ( 42 ). Previous studies suggested that osteoblasts were the main source of OPG.…”

“…The ability of macrophages to phagocytose endows them with a variety of tissue‐specific trophic roles. Osteoclasts, for example, are bone‐resident macrophages that destroy senescent bone cells and, as such, regulate bone morphogenesis . Metchnikoff also performed the first intravital imaging of macrophages, taking advantage of the transparency of starfish larvae.…”

“…144 Collective data suggest that early uNK cells along with monocytes and trophoblasts are major sources of ANGPTs to regulate decidual spiral artery modeling and normal placentation. 145,146 ANGPT1 and ANGPT2 are detected in human uNK cell culture supernatants and both are at lower levels in second than in first trimester samples. 57 Increases in ANGPT1, ANGPT2 and IFNG (at 10 ng/ml level) are proposed to disrupt human vascular smooth muscle cells and breakdown extracellular matrix components 63 to enhance early angiogenesis.…”
