Immuno-spin trapping detection of antioxidant/pro-oxidant properties of zinc or selenium on DNA and protein radical formation via hydrogen peroxide

Abstract: Trace elements can participate in the catalysis of group-transfer reactions and can serve as their structural components. However, most of them including zinc and selenium have multifunctional roles in biological environments such as antioxidant and/or pro-oxidant effects, as concentration-dependent manner. Although it has been demonstrated the antioxidant actions of either selenium or zinc compounds, there are several documents pointing out their pro-oxidant/oxidant roles in biological systems. Here we have u… Show more

“…The possible mechanisms of protective role of sodium selenite might be associated with following factors: (1) Se was the nucleus of antioxidant selenoenzymes which could alleviate oxidative-stress-associated kidney injury [ 42 ]. Indeed, AFB 1 -induced tissue injury was mediated through oxidative reaction [ 28 ]; (2) Se could reduce the formation of DNA adducts of aflatoxin in the chick [ 43 , 44 ]; and (3) Se could attenuate the decreased activity of Na + -K + ATPase induced by AFB 1 . Our result showed that Se could elevate AFB 1 -depressed Na + -K + ATPase activity which was accordance with the recent research [ 45 ].…”

“…The pioneering work for the development of IST was performed by the research group of Mason et In principle, all macromolecular radicals, derived from e.g., proteins or DNA, can be measured very specifically with IST. 451,[462][463][464][465][466][467][468][469] A novel and intriguing extension of the IST methodology arose from a combination with molecular magnetic resonance imaging (mMRI) for in-vivo and in-situ localization of radical sites in preclinical studies. In this case, chelated paramagnetic gadolinium (Gd), a well-known MRI contrast agent, is linked to a modified anti-DMPO.…”

“…A number of previous studies have shown that zinc is required for many physiological processes including growth, neuronal function, reproduction, and immune system (5). Zinc deficiency, in this regard, has been implicated in a wide range of diseases, including immune system disorders, lymphopenia, intrinsic immunity, and increased infectious diseases (8,9). Selenium is the other essential element demanded for the body.…”
