“…We confirmed earlier reports showing the beneficial effects of enzyme pretreatment on tissue antigenicity of NBF-fixed sections (Huang et al, 1976;Denk et al, 1977;Witting 1977;Brozman, 1978;Qualman and Keren, 1979;Swoveland and Johnson, 1979;Johnson et al, 1980;Hondo et al, 1981). We have not seen any reports describing enzyme treatment of virus infected chicken tissues.…”

“…Our trypsin treatment was efficient in unmasking IBDV and IBV antigens although parts of tissues did not always adhere to the glass slides. Pronase digestion was carried out according to Witting (1977) and was shown to be especially efficient in revealing IBDV antigen. The epithelial and connective tissue components of trachea were damaged by pronase treatment, making identification of IBV impossible.…”

“…Sections were exposed for 90 min to 50 μM BdABK. Thereafter, slides were washed twice (1 min) in PIPES and fixed with 4% para-formaldehyde [ 36 ]. Slides were washed three times (5 min) and then exposed to 1 M of glycine for 90 min to eliminate autofluorescence from aldehyde-fixed tissue.…”
