Immunogenic sonodynamic therapy for inducing immunogenic cell death and activating antitumor immunity

Abstract: Immunotherapy is widely regarded as a promising treatment for cancer. However, the immune effector phase suppression of tumor microenvironment (TME) and the generation of immune-related adverse events limit its application. Research indicates that sonodynamic therapy (SDT) can effectively activate antitumor immunity while killing tumor cells. SDT produces cytotoxic substances of tumors, and then cell apoptosis and immunogenic death occur by selectively activating the sonosensitizer under ultrasound. In recent … Show more

“…Taking into account the inherent immunostimulatory properties of bacteria and the potential of SDT in activating immune responses, [ 50,51 ] we conducted a thorough investigation to elucidate the underlying mechanisms responsible for the remarkably enhanced therapeutic effect achieved by E. coli ‐pE@PCN‐mediated SDT. Mice bearing CT26 tumors were randomly assigned to five groups and subjected to identical treatments as described above.…”

“…SDT effectively eradicates tumor cells, triggering the induction and release of tumorassociated antigens (TAAs) and generating damage-associated molecular patterns (DAMPs). These DAMPs include calreticulin (CRT) exposed on the cell surface, high-mobility group protein 1 (HMGB1) secreted from the tumor cells, and ATP and heat shock proteins (HSP70, HSP90) released by the cells [29]. In traditional antitumor therapies such as chemotherapy or radiotherapy, apoptosis in tumor cells often leads to the release of DAMPs.…”
