Impact of Eurycoma longifolia extract on DNA integrity, lipid peroxidation, and functional parameters in chilled and cryopreserved bull sperm

Abstract: This study aims to assess the effect of Eurycoma longifolia aqueous extract on chilled and cryopreserved quality of bull sperm. Semen samples were obtained from four Simmental-Brangus. Each sample was divided into two fractions: the first fraction was used for chilling the semen, and the second fraction was used for the freezing process. Both fractions were extended with Tris-egg yolk extender supplemented with 0.0, 0.25, 0.5, 1.0, 2.5, 5.0, and 7.5 mg/ml Eurycoma longifolia aqueous extract. The diluted chille… Show more

“…Then, the preputial orifice was washed with a saline solution and dried with a paper towel. Rectal evacuation was performed before inserting the probe and switching on the electroejaculator device, whose voltage was increased automatically; then, the ejaculate was collected in a sterile plastic sleeve (Baiee et al, ). Sperm concentration and the total motility of the ejaculate were assessed using a spectrophotometer (Spermacue ® ) and an optical microscope at 400× magnification (Eclipse E200 ® , Nikon Inc), respectively.…”

“…Semen preserved in liquid or frozen form are common procedures in inseminating cows; roughly 95% of AI dosages are in the form of frozen semen straws (Amann and Waberski, 2014). Nevertheless, freezing-thawing practices of prolonged semen are accountable for a substantial decrease in motility, viability, membrane integrity and morphology of sperm (Amirat-Briand et al, 2010;Yimer et al, 2014;Baiee et al, 2018a) resulting in decreased rate of fertility of the frozen semen (Büyükleblebici et al, 2014). In actual fact, chilled semen possesses a high rate of fertility in comparison to frozen semen (Medeiros et al, 2002;Galarza 2019); nonetheless, chilled semen can be kept for a few days only (Papa et al, 2015).…”

“…Subsequently, the preputial orifice was washed with saline solution and dried with a paper towel. Following this, rectal evacuation was performed before inserting the probe and switching on the electroejaculator device, whose voltage was increased automatically (Baiee et al, 2018). Subsequently, the ejaculate was collected in a sterile plastic sleeve and its color, volume, pH, sperm concentration, and mass motility were determined using a graduated glass collection tube, pH-indicator strips (Merck, Darmstadt, Germany), a specific spectrophotometer (Spermacue ® , Minitube, St Louis, MO, USA) and a phase contrast microscope (100× magnification) (Domínguez et al, 2008), respectively.…”

“…A minimum of spermatozoa were immediately analyzed in different fields (400×; Eclipse E200, Nikon Inc., Tokio, Japan). Reacting spermatozoa (rolled tails) were considered as sperm with intact membranes (Baiee et al, 2018). The percentage of reacting spermatozoa was calculated.…”
