Impedimetric DNA Aptasensor for Sensitive Detection of Ochratoxin A in Food

Abstract: We report a high sensitive biosensor based on DNA aptamers for detection of ochratoxin A (OTA). The thiolated DNA aptamers specific to OTA of different configurations have been immobilized by chemisorption to the surface of a gold electrode. Electrochemical impedance spectroscopy (EIS) in the presence of a redox probe [Fe(CN) has been used for the determination of the charge transfer resistance, R ct , followed by addition of OTA. The R ct increased with increasing OTA concentration in the range of 0.1-100 nM… Show more

“…These steps add time, and this is one reason for the continued development of non-enzymatic labels and label-free devices in order to achieve portability of the devices [47,48]. These disadvantages can be also overcome with the use of aptamer nucleotide sequences and AuNPs labels that are able to discriminate between various toxin categories [42][43][44][45]. However, for the time being, such formats tend to require expensive, laboratory-based equipment that requires significant technical training to operate.…”

“…Additionally, an impedimetric DNA aptamer biosensor onto a gold electrode's surface has also been reported [45]. EIS has been used for the determination of the charge transfer resistance (R ct ) onto a gold electrode's surface, where thiolated DNA OTA aptamers of different configurations have been immobilized by chemisorption.…”

“…The system was further improved [87], showing a detection limit comparable to that of the HPLC method (0.12 nM), and was validated in food samples. Another design for the OTA detection proposed the use of a long polyethylene glycol spacer chain, which led to the formation of long tunnels at the surface of screen-printed carbon electrodes, with aptamers acting as gates.…”

“…Many different procedures have been used to improve the sensitivity of the G-quadruplex electrochemical aptasensors using amplification strategies based on redox labels, such as ferrocene [72][73][74][75][76][77][78][79][80] A c c e p t e d M a n u s c r i p t 13 transducers have been employed, such as modified platinum [89] or carbon electrodes [64,76,[89][90][91][92].…”
