Improving 2-Phenylethanol Production via Ehrlich Pathway Using Genetic Engineered Saccharomyces cerevisiae Strains

Abstract: 2-phenylethanol (2-PE) is an important aromatic compound with a rose-like fragrance widely used in food industry and cosmetic manufacture. In order to obtain "natural" 2-PE, the genetically modified budding yeasts were developed and applied for the 2-PE production. The gene ARO8 encoding transaminase and the gene ARO10 encoding decarboxylase in the Ehrlich pathway were expressed in Saccharomyces cerevisiae S288c. The activities of transaminase and decarboxylase were both enhanced in the corresponding recombina… Show more

“…It is noteworthy that the production rate achieved with AM1-d ( P = 53.19 mg of 2-PE/L/h) is even higher than the rate recently reported for the genetically engineered S. cerevisiae SPO810 strain, which produced 2.61 g/L of 2-PE after 60 h ( P = 43.50 mg of 2-PE/L/h); however, this was in a fed-batch process [Yin et al, 2015]. While in a batch culture in a shaking flask the SPO810 strain produced only 1.10 g/L of 2-PE.…”

“…Recent reports show that both de novo synthesis and the production of 2-PE via L -Phe biotransformation can be significantly increased by applying metabolic engineering [B. ; T.-Y. Romagnoli et al, 2015;Yin et al, 2015]. However, because of complicated legislation and negative consumer attitude to genetically modified organisms (GMO), non-GMO strategies are highly recommended and have previously been successfully applied to industrial yeast strains, including the improvement of ethanol tolerance [Stanley et al, 2010], copper resistance [Adamo et al, 2012], or aroma production [Steensels et al, 2014].…”

“…The presence of specific nitrogen sources, including ammonia, may regulate both the Ehrlich and Shikimate pathways in a number of places. For instance, ARO8 and ARO10 are two key genes in the Ehrlich pathway and their expression has been shown to influence 2PE production30 and to be repressed by ammonia, resulting in lower 2PE production 29…”

“…Another strategy is transamination of l -phenylalanine to phenylpyruvate by aminotransferase. Several groups have demonstrated the biosynthesis of 2-phenylethanol from l -phenylalanine with a high titer by transamination (Kim et al 2014; Yin et al 2015). …”
