Introduction: Medicinal plants are chief antidotes for numerous diseases and have been used since time immemorial. Sweet flag’s (Acorus calamus) presence is in Ayurveda and belongs to the genus Acorus L. of the family Acoraceae and is widely distributed in temperate to sub temperate regions. It is commonly used to treat appetite loss, diarrhoea, digestive disorders in traditional medicinal systems of Asian and European countries. The aim of this study is to explore the phytoconstituents, antioxidant and anti-inflammatory potential of methanolic root extract of Acorus calamus. Materials and Methods: Methanolic root extract of Acorus calamus was done by the Hot Percolation method. Later it was dried and used to analyse the antioxidant and anti-inflammatory potentials. Phytochemical screening was done to analyse the presence of various phytochemicals. Antioxidant effect of Acorus calamus was tested by 2, 2-Diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity and Albumin denaturation inhibitory potential test was organised for testing it’s anti-inflammatory Activity. The data were analysed statistically by a one-way analysis of Variance (ANOVA) followed by Duncan’s multiple range test was used to see the statistical significance among the groups. The results with the p<0.05 level were considered to be statistically significant. Results: Methanolic root extract of Acorus calamus was found to be rich in Alkaloids, Flavonoids, Terpenoids, Sapanoids, Steroids and Phlobatannin. The presence of phytochemicals like alkaloids, saponins, Flavonoids indicates that the extract has potential for further in vitro analysis like antioxidant and anti-inflammatory potentials. It was observed that Acorus calamus has both antioxidant (IC50 of = 295 µg/ml) as well as anti inflammatory potentials (IC50 =310 µg/ml) and the activity increased in a dose dependent manner as compared to that of standard (Vitamin C and Diclofenac respectively). Conclusion: The study proves the antioxidant and anti-inflammatory efficacy of Acorus calamus and throws light on the prospects of drug formulation against oxidant activity and inflammation.

Medicinal plants have been treating various ailments and diseases since ancient times. Aquatic and semiaquatic medicinal plants play an essential role in human welfare to fulfill their daily needs. They have shown biological, pharmacological, nutraceutical, and commercial applications. This review aims to collect and update all recent information on ethnomedicinal, phytochemistry, pharmacological activities, and nanoparticle synthesis and their uses in aquatic and semiaquatic medicinal plants. Original research papers, review papers, short communications, and book chapters on aquatic and semiaquatic plants have been retrieved from PubMed, Web of Science, Scopus, and Google Scholar. Keywords, ethnomedicinal studies, phytochemistry, pharmacological activities, and nanoparticle synthesis from aquatic and semiaquatic medicinal plants are used for the search. Different aquatic and semiaquatic medicinal plants belonging to the families Acanthaceae, Alismataceae, Amaranthaceae, Apiaceae, Araceae, Asteraceae, Boraginaceae, Ceratophyllaceae, Cyperaceae, Fabaceae, Hydrocharitaceae, Lythraceae, Marsileaceae, Menyanthaceae, Nelumbonaceae, Nymphaeaceae, Onagraceae, Plantaginaceae, Poaceae, Polygonaceae, Pontederiaceae, Primulaceae, Scrophulariaceae, and Zingiberaceae have been studied. They are rich in alkaloids, flavonoids, terpenoids, phenolics, saponins, tannins, dietary fiber, glycosidic derivatives, carbohydrates, and proteins. These phytochemicals have been used for their antimicrobial, antioxidant, hepatoprotective, sedative, anticonvulsant, cytotoxic, antiparasitic, and antidiabetic activities. Besides this, various parts of the plants are used as dietary supplements and green nanoparticle synthesis. These plants are also known for their commercial value and are used as an ingredient in some pharmaceutical industries.

The essential oil (EO) of the rhizome of Acorus calamus Linn. was isolated by using a Clevenger apparatus and extracts were prepared by cold percolation technique using the solvents hexane and methane. The chemical constituents of EO were analyzed by Gas Chromatography-Mass Spectroscopy (GC-MS) analysis. A total number of nine chemical compounds were identified and quantified occupying 100% of the total oil composition. The major chemical constituent was reported to be β-asarone (84.87%). Acid value, saponification value, and iodine number of the oil were measured and found to be 0.24 mg KOH/g, 0.42 mg KOH/g, and 31.75 g I2/100gm, respectively. The antibacterial activity of the hexane and methanol extract was examined against two bacteria by the agar well diffusion method. The hexane extract showed antibacterial activity against E. coli with a zone of inhibition(ZOI)of 10 mm, and. subtilis with ZOI of 7 mm. The methanol extract showed antibacterial activity against B. subtilis only, with a ZOI of 4 mm. Hexane and methanol extract also showed significant antifungal activity against fungi C. albicans with a ZOI of 6 mm and 5 mm, respectively.DPPH assay showed that the percentage of free radical scavenging activity increased with an increase in the concentration of the extract. The total phenolic content of the methanol extract of A. calamus was found to be 48.36 mg/g GAE.
