The effects of carbohydrate availability, carbohydrate source, nutrient availability and illumination on germination and early development of Bletia purpurea (Orchidaceae) seeds were investigated using asymbiotic seed germination. Of special interest was determining the minimum nutritional and light requirements for the completion of germination. Germination and development was limited when seeds were cultured in darkness without sucrose. Seeds were able to germinate under illuminated conditions even in the absence of sucrose and this effect was enhanced when mineral nutrients were incorporated into media. Sucrose, fructose, glucose and trehalose enhanced germination and seedling development while mannitol and sorbitol did not. These data suggest that carbohydrates, either as products of photosynthesis, from symbiotic fungi in situ or as exogenously supplied sugars in vitro, play an important role in regulating seed germination by fulfilling an energy requirement. This hypothesis has been often expressed but rarely satisfactorily tested. Mineral nutrients appear to be less important for germination than carbohydrates. The differential effect of sucrose, fructose, glucose and trehalose at two different concentrations on rhizoid production indicates carbohydrates may play a role in regulating rhizoid production.

The effects of explant and cytokinin types on in vitro plant regeneration of Ansellia africana were investigated. The exogenous addition of cytokinins is not required for the proliferation of new protocorms from Trimmed protocorm cluster (TPC) explants. To the contrary, nodal and shoot-tip explants produced a single shoot in response to the addition of cytokinins. Overall plant growth in terms of shoot length, leaf number, frequency of root organogenesis, root length, and fresh weight/plant were significantly higher in media containing meta-Topolin Riboside (mTR) in both nodal and shoot-tip explants. Thidiazuron (TDZ) and 6-benzyladenine (BA) induced stunted and hypertrophied shoots at their highest level (15 lM). In addition root differentiation and root growth were significantly lower on P668 media with TDZ and BA. Zeatin was capable of inducing a significantly higher root organogenesis frequency and root length in TPC explants as compared to other cytokinins. However, TPC explants produced a significantly greater number of longer shoots ([3 cm) on P668 media with mTR. Hyperhydric shoots were produced from TPC explants. Vanda tricolor Lindl. Var. Suavis forma Bali has a specific character than the forma Merapi and forma West Java that is size of flowers and fruits, spots purple in labellum purple colored labelum and floral fragrance. High genetic diversity is one of the major factors in breeding improvement. Increasing the diversity of orchid plant characteristics can be done by giving paclobutrazol. Treatment by paklobutrazol in culture medium aims to inhibit growth both in number of leaves and shoot length so that the plant becomes smaller size than their parent or original. The result of growth resistance that occurs varies based on the concentration of paclobutrazol added to medium Vacint & Went (VW) in culture of seeds V. tricolor orchid added with coconut water with concentration of paklobutrazol Control K0 (0%), K1 (1mg / l), K2 (3mg / l), K3 (5mg / L), and K4 (7mg / l). Each treatment performed 5 times repetition. The result of planting of Vanda tricolor seed explants with paclobutrazol modification in vitro was found 40% cultured imbibition and then protocorm, 8% browning on seed, and 52% contamination. This study showed descriptively the orchid seed Vanda tricolor responded to Vacint and Went (VW) media which was added with plant growth regulator of paklobutrazol, so that it was able to grow and develop until reaching phase 1 that seeds to form protocorm, but statistically the addition of plant growth regulator of paklobutrazol has not show a significant influence on the growth and growth response of orchids Vanda tricolor (P> 0.05) because the observation time is not sufficient for at least 6 months of observation and see the function of paklobutrazol is to slow the growth.
