In vitro development of human primordial follicles to preantral stage after vitrification

Abstract: Purpose The aim was to culture primordial follicles in vitro to reach preantral stage in vitrified human ovarian tissue. Methods Ovarian tissue samples were obtained from six women. Tissue strips were vitrified by infiltration with a cryoprotectant followed by mounting on a stainless steel carrier. After culturing for 7 days the morphology and developmental stages of follicles enclosed in fresh and vitrified groups were analyzed. Results High proportion of viable follicles in vitrified ovarian strips was obtai… Show more

“…Usually, a short period of 2 days is not enough to assess changes in the follicular density of follicular growth [38,39]. However, the growth of follicles during the 7-day culture that we observed was in accordance with previous studies [40].…”

“…In vitro culture of ovarian follicle is a valuable bioengineering technique for investigating the factors that control oocyte development at different developmental stages and as an alternative therapy for fertility preservation (Khosravi et al , ; Kim et al , ). However, currently available in vitro follicle culture techniques are labour‐intensive and of low efficiency, as most investigators collect and seed single pre‐antral follicles for monofollicular maturation.…”

“…Vitrification of ovarian tissue is a promising approach with great potential to preserve fertility (Donnez et al. ; Donnez et al, ; Khosravi et al., ) of non‐human animals; however, few data are available, especially for canids. To our knowledge, this study was the first to assess follicular density and histological features from fresh and vitrified–warmed ovarian cortex from domestic dogs.…”

“…Isolated follicles in a 3D matrix have been cultured in vitro in an attempt to obtain antral follicles. Good results were obtained for primates (Khosravi et al., ; Xu et al., ) and rats (Wang et al., ); however, follicle isolation in dogs has been proven to be laborious and with a low rate of success (Ackermann et al. ).…”
