A stable and biocompatible targeting complex CFNs@PEG-FA is developed. The initially synthesized cobalt ferrite nanoparticles (CFNs) were treated with poly(ethylene glycol) (PEG) in order to improve biocompatibility of the CFNs. Citric acid (CA) was used as the coupling agent, which made PEG to bond with the CFNs. CFNs@PEG were conjugated with folic acid (FA) to synthesize CFNs@PEG-FA, which was capable of targeting the FA receptor positive (FAR+) cancer cells. Synthesized nanoparticles were physically and chemically analyzed using EDX, FT-IR, XRD, TGA, FESEM, TEM, DLS, and VSM. The biocompatibility of CFNs@PEG-FA was assessed in vitro on HSF 1184 (human skin fibroblast cells) and HeLa (human cervical cancer cell, FAR+) using MTT assay and AO/EB staining florescence method. High level of CFNs@PEG-FA binding to HeLa was confirmed through quantitative and qualitative in vitro targeting studies. Results show that CFNs@PEG-FA can be a potential biomaterial for use in biomedical trials, especially magnetic hyperthermia. The findings through this in vitro study are to be compared in future with those of in vivo studies.

Nanotechnology is a practical and exquisite method for enhancing the bioavailability of insect medicines. The present work deals with nano-encapsulation of freeze-dried Polyrhachis sp. (Pol) to increase its aqueous solubility and bioactivity. In the current study, Polyrhachis sp. was encapsulated into carbonate calcium (CaCO 3 ) through a one-step co-precipitation method to produce CaCO 3 @Pol nanoparticles (NPs). It was characterized accordingly by FTIR, FESEM, DLS, and zeta potential analysis methods. Null CaCO 3 and CaCO 3 @Pol particle sizes were observed in the nano range, and most of the nanoparticles had spherical shapes. The cytotoxicity evaluation of neat Pol and CaCO 3 @Pol was performed in vitro on human breast cancer cells (MCF7) and human gastric cancer cells (AGS) through an MTT assay. Morphological alternation studies disclosed that CaCO 3 @Pol-treated cells had undergone apoptosis since shrinkage and apoptotic bodies were observed. The half-maximal inhibitory and 50% lethal dose of neat Pol and CaCO 3 @Pol were calculated. Besides, the RT-PCR technique analyzed the status of apoptosis genes such as caspase-3, caspase-8, caspase-9, Bcl-2, and Bax expressions. The molecular mechanisms provoked apoptosis through modulating pro (caspase-8, caspase-9, Bax) and anti-apoptotic (Bcl-2) mediator genes expression in AGS cells. The findings conveyed that the CaCO 3 @Pol is a promising biomaterial for future biomedical applications.

Paraquat (PQ) is a commercially important and effective herbicide in the world. Nevertheless, it has higher toxicity causing acute organ damage and different complications, mainly in the lungs and kidneys. Ferulic acid (FA), 4‐hydroxy‐3‐methoxycinnamic acid imposes multiple pharmacological impacts. No protective effect of FA on PQ poisoning‐caused human embryonic lung fibroblast damage has not been reported. Despite their many beneficial effects, FA is characterized by poor water solubility, low bioavailability, and phytochemical instability. To solve the problem, β‐cyclodextrin nanosponge (β‐CD NSs) was utilized to increase the solubility of FA so that it was grafted into β‐CD NSs to establish β‐CD@FA NSs. The purpose of this work was to examine for the first time the protective effect of β‐CD@FA NS on MRC‐5 human lung cells damages induced by PQ poisoning. MTS assay was performed to investigate the viability of MRC‐5 cells at different concentrations of FA/β‐CD@FA NSs when cells were co‐cultured with 0.2 μg/mL PQ. The flow cytometry study was carried out to determine apoptosis. Malondialdehyde (MDA), superoxide dismutase (SOD), and catalase (CAT) levels were detected using appropriate biochemistry kits. Compared with the PQ group, the cell activity, CAT, and SOD levels were significantly increased in the FA and chiefly in β‐CD@FA NSs intervention groups, whereas apoptosis and MDA levels were markedly decreased. The inflammatory factors tumor necrosis factor‐alpha (TNF‐α), interleukin 6 (IL‐6), and interleukin 22 (IL‐22) were detected. The results demonstrate that β‐CD@FA NSs can inhibit PQ‐induced cell damage by enhancing antioxidant stress capacity and regulation of inflammatory responses.
