In Vitro Lung Model Assessing the Efficacy of Surgical Sealants in Treating Alveolar Air Leaks

Abstract: TachoSil demonstrated a strong sealing efficiency and marked elasticity in treating AAL. These results are consistent to that of the previously published animal experiment, suggesting the reliability of the presented in vitro model.

“…On the other hand, they are often performed in nonphysiological conditions like room temperature and the measured endpoint is often shortterm maximal burst pressure, which might not be the most clinically relevant quality. [9][10][11][12][13][14][15]21 Only 2 studies evaluated the mechanical resistance over a longer period than a few minutes. 22,23 Two other studies evaluated the biochemical 24 and the structural effects of a long-term incubation with biological fluids.…”

“…19,20,30 Several experiments already evaluated the maximal burst pressure of TachoSil applied on different biological membranes under room conditions. [10][11][12]14,31,32 Another experiment included an in vitro long-term observation of the biochemical stability of TachoSil in PF by measuring d-dimers and hydroxyproline levels, which are degradation products of TachoSil. This experiment showed a significant higher breakdown of TachoSil in PF compared with saline, IF, and PF combined with collagenase inhibitor.…”
