In vitro Pb exposure disturbs the balance between Aβ production and elimination: The role of AβPP and neprilysin

Abstract: Metabolism of β-amyloid peptide (Aβ) is closely associated with the pathology and etiology of Alzheimer’s disease (AD). Our previous studies on aging primates and rodents have revealed that early life lead exposure increases the expression of the β-amyloid precursor protein (AβPP), elevates Aβ levels, and promotes neurodegeneration in old age. These effects were attributed to de novo synthetic pathways; however, the impact on Aβ degradation was not explored. Neprilysin (NEP), a rate-limiting catabolic peptidas… Show more

“…SH-SY5Y cells were seeded in a white-walled clear bottom 96-well plate (3000 cells/well) and incubated for 24h in a humidified 37°C incubator with 5% CO 2 . Cell differentiation was induced by treating SH-SY5Y cells with 10 µM all- trans -retinoic acid (RA) for 3 days following established procedures [40,41]. Cell count was performed in randomly selected wells and the mean value was calculated.…”

“…High levels of Pb in bone were found to be associated with poor cognitive function in elderly workers in one study [6] although serum Pb levels had no correlation with dementia in another [7]. Nonetheless, a recent study suggests Pb may increase APP levels in cells [23]. The role of Cadmium in development of AD is not established.…”

“…On the other hand, an overloading of the main clearance mechanisms of this peptide from the brain and an increase of its influx into the CNS may also lead to Ab accumulation, aggregation, and neurotoxicity. Among many Ab peptide-degrading enzymes, neprilysin, a membrane-bound zinc metallopeptidase, is considered the most important Ab peptide-degrading enzyme (Huang et al, 2011;Iwata et al, 2001). The neprilysin is capable of breaking down Ab in both intra-and extra-cellular spaces via its active site at the lumen side.…”
