Objective: The aim of the present investigation was to isolate and identify Lactobacillus strains from dairy and cattle dung samples. Potent isolates were selected for screening by antimicrobial activity; selected lactobacilli were further tested for probiotic properties and adhesive attributes.Methods: Lactobacilli were isolated aseptically on specific de man, rogosa and sharpe medium from dairy and cattle dung samples. Isolates were identified by Gram-staining, motility, catalase, endospore, and carbohydrate fermentation tests. Further, the isolates were screened for antimicrobial activity by disk diffusion assay, and potent lactobacilli were observed for probiotic properties: Acid and bile salt tolerance, gelatinase activity, and autolytic activity. For analyzing the adhesive attributes, isolates were observed for autoaggregation, coaggregation and microbial adhesion to solvents assay. Results: About 12Lactobacillus strains among 98 isolates exhibited maximum antimicrobial activity were further selected for identifying their probiotic and adhesive attributes. Among 12 selected isolates, cell-free supernatant (CFS) of buffalo milk BM10 and goat milk GM10 showed excellent antimicrobial activity, 20.34±0.02 mm against Staphylococcus aureus and 18.65±0.11 mm against Escherichia coli. Isolates showed survival at pH 2 and 3 and can tolerate 0.2-0.3% bile salt concentrations. The GM5 showed maximum autoaggregation (67.04±0.61%) and minimum coaggregation (11.51±0.50%) showed by GM3. The BM10 exhibited maximum adherent value 64.84±1.41% for n-hexadecane. Conclusion:The two lactobacilli, BM10 and GM10 identified as Lactobacillus fermentum and Lactobacillus pentosus on the basis of phenotypic and sugar utilization tests. The CFS of both lactobacilli can be used as antimicrobial agent. Both isolates showed significant results of probiotic and adhesive attributes, therefore, can be evaluated for clinical and therapeutic applications.

Objective: The objective of this study was to identify the potential yeast isolates at themolecular level and to evaluate their probiotic characteristics.Methods: Molecular characterization was done for five potential probiotic yeast strains. In vitro assays have been conducted to evaluate the probiotic properties such as NaCl tolerance, autoaggregation, and coaggregation. Hemolytic activity, urease activity, and cytotoxicity tests were carried out for safety assay during the characterization of yeast strains. Results:In this study, the yeast strains, viz., LM, MR, GOI, GII2, and WI was identified at the molecular level and named as Yarrowia lipolytica VIT-MN01, Kluyveromyces lactis VIT-MN02, Lipomyces starkeyi VIT-MN03, Saccharomycopsis fibuligera VIT-MN04, Brettanomyces custersianus VIT-MN05, respectively. Maximum autoaggregation (92%) and coaggregation (97 %) were noted in case of L. starkeyi VIT-MN03. All yeast strains showed nonhemolytic activity. In vitro toxicity assay was performed and all the yeast strains showed nontoxic nature. Conclusion:Five yeast strains have been studied for their probiotic characteristics and identified at molecular level. Out of five yeast strains, three strains showed maximum adhesion ability, which is a prerequisite for colonization and protection of gastrointestinal tract. All the yeast strains are validated as a safe bioresources because of their nonhemolytic activities and nonproduction of urease. It can be concluded that the identified yeast strains can serve as promising probiotics in various fields of food industry.

ABSTRACT:The studies in the present article aims at developing an optimal oral polyherbal nanoemulsions from Carissa carandas (CC) and Carissa spinarum (CS) which are noticed in ethnobotanical herbarium for their pharmacological relevance. Nanoemulsions were developed in different proportions of oil and surfactant systems in order to enhance its oral bioavailability. Solubility studies were carried out for the preparation of the formulation. Pseudoternary phase diagrams were constructed by aqueous titration technique and different polyherbal nanoemulsions were prepared. Formulations selected from o/w nanoemulsion region were subjected to various thermodynamic stability tests. Optimized formulations were evaluated for physicochemical tests. In vitro anti-inflammatory activity was carried out by inhibiting heat induced albumin denaturation, proteinase activity, red blood cells membrane stabilization and broth dilution assay. Phase diagram with 1:2 S/Smixhas shown largest area of nanoemulsion. In vitro dissolution studies revealed that release of drug from nanoemulsion 2 (96.88±1.29%) was faster than the nanoemulsion1 (85.36±2.12%) in 120 min. The formulation showed satisfactory results for in vitro anti-inflammatory activity by inhibiting the heat induced albumin denaturation and red blood cells membrane stabilization as well as significant inhibition for proteinase activity at 600μg/ml. The NE2 showed excellent antibacterial activity against Staphylococcus aureus, Bacillus subtilis and Salmonella typhi whereas NE1 against Escherichia coli. These studies indicate that the nanoemulsion may be used as a promising drug delivery vehicle for bioavailability enhancement of hydrophobic drugs.
