Increase of basic fibroblast growth factor (FGF) and FGF receptor messenger RNA during rat thyroid hyperplasia: temporal changes and cellular distribution

Abstract: Goitre was induced in adult rats by acute (1 or 2 weeks) or chronic (4 or 10 weeks) administration of methimazole together with a low iodine diet. Involution of thyroid growth was then observed at 16 weeks, 4 weeks after withdrawal of goitrogens and reversion to a normal diet. Experimental animals quickly became hypothyroid compared with controls and exhibited thyroid hyperplasia (control (n = 10): total serum thyroxine (T4) 66 +/- 4 nmol/l, thyroid weight 5 +/- 1 mg/100 g body weight, means +/- S.D.; experime… Show more

“…Basic FGF (fibroblast growth factor) is present in human thyroid tissue (48) and there are stores of FGF in the basement membrane of follicles in normal adult rat thyroid (49). Moreover, bFGF expression increases during thyroid hyperplasia in rodents (50).…”

“…RNase protection assays were performed with a Guardian kit (Ambion) 12 with the bFGF RNA probe prepared by in vitro transcription from a bFGF cDNA insert previously cloned in the TA vector (Invitrogen). An RT-PCR product from BAECs using bFGF primers as above was inserted into the TA cloning vector and completely sequenced.…”

“…In thyroid, increased expression of FGFR1 has been observed in benign and malignant tumors (4,8). FGFR1 is up-regulated in rat thyroid follicular cells upon goitrogen administration, whereas a dominant-negative FGFR1 reduces goitrogenesis in mice (9,10).…”
