Increased Expression of PcG Protein YY1 Negatively Regulates B Cell Development while Allowing Accumulation of Myeloid Cells and LT-HSC Cells

Abstract: Ying Yang 1 (YY1) is a multifunctional Polycomb Group (PcG) transcription factor that binds to multiple enhancer binding sites in the immunoglobulin (Ig) loci and plays vital roles in early B cell development. PcG proteins have important functions in hematopoietic stem cell renewal and YY1 is the only mammalian PcG protein with DNA binding specificity. Conditional knock-out of YY1 in the mouse B cell lineage results in arrest at the pro-B cell stage, and dosage effects have been observed at various YY1 express… Show more

“…In this regard, YY1 expression is elevated in various types of cancer (25). Although numerous studies have been devoted to understanding the roles of YY1 in B cell development and V(D)J recombination of the Igh and Igk loci (18, 21, 2629), studies of YY1 in T-lineage cells have been limited to its role in regulating Th2 cytokine production (30). …”

“…YY1 has since been implicated in a number of processes, including development of muscle (25)(26)(27)(28)(29)(30) and B cells (31)(32)(33), and stem cell regulation (34)(35)(36)(37). In embryonic stem cells, YY1 is part of the Myc transcriptional network (36) but counterintuitively, suppresses induced pluriopotent cell reprogramming (34).…”

“…In embryonic stem cells, YY1 is part of the Myc transcriptional network (36) but counterintuitively, suppresses induced pluriopotent cell reprogramming (34). In the blood, YY1 overexpression promotes long-term hematopoietic stem cell maintenance (37). Conversely, YY1 promotes satellite cell activation and differentiation during muscle regeneration (35).…”

“…The expression of specific TFs selected for their binding to the CD34 promoter, their involvement in NO signaling in various systems, and their importance in the hematopoietic system was examined in NO-treated HSCs from juvenile and adult mice. Yy1 , a multifunctional polycomb group (PcG) transcription factor, has been shown to mark phenotypically defined and functional LT-HSCs and is also known to interfere with the commitment process [30]. Consistent with these reports, Yy1 was found to be significantly upregulated by NO in juvenile HSCs, but not in adult HSCs.…”

“…7 h). Also, the genes related to self-renewal such as Yy1 , Chop1 , and Runx1 [3032] were significantly upregulated in juvenile HSCs as compared to the adult HSCs. Sp1 is known to positively regulate the CD34 promoter [33].…”
