Increased interleukin‐6 production during the acute phase of the syndrome of episodic angioedema and hypereosinophilia

Abstract: The plasma histamine level was within normal range. The level of IL-6 in sera peaked to 74 pg/mL, concomitant with the peak of eosinophilia at the acute phase phase of the attack. Under corticosteroids, we observed a drop in the IL-6 serum level to 29 pg/mL, concomitant with the clinical remission. During the attack, an increase in IL-6 production was observed in 24 h blood monocyte supernatants (11.10(3) pg/mL compared with 2.4+/-0.8.10(3) pg/mL for BM from controls) as well as in skin endothelial cells but n… Show more

“…IL-6 is produced by a wide variety of cell types and is known to contribute to Th2 cytokine production, lung eosinophilia, and airway hyperresponsiveness [ 32 ]. As such, it was found to be higher in serum of asthmatic patients [ 33 , 34 ] and mice [ 32 ]. In our study, as expected, IL-6 was significantly higher in lung homogenates from ova-ova WT group compared to the sal-ova controls.…”

“…The angioedema of EAE and NEAE is likely associated with a temporal increase in the vascular permeability of local capillaries via some mediators, including antiendothelial cell antibodies and cytokines3, 14-16). Several reports have shown that histamine levels are within normal limits, and that IL-1, IL-5, and IL-6 generated by monocytes, endothelial cells, T cells, and eosinophils increase during the acute phase of EAE3, 5, 11, 15, 16). According to a recent report, IL-5 levels were determined to be lower, and TNF-αlevels were higher in the acute phase of NEAE than in the acute phase of EAE.…”

“…Many other cases have been detected since this initial report, and this syndrome has been recognized as a distinct clinical entity2, 3). More than 40 cases with symptomatic presentations and laboratory findings similar to EAE have been reported, but these cases have not involved recurrent attacks, and thus have been subsequently designated as nonepisodic angioedema with eosinophilia (NEAE)4-8).…”

“…Eosinophil activation is also associated with acute symptoms such as flare‐ups (Tillie‐Leblond et al , 1998; Vandezande et al , 1999; Daneshpouy et al , 2002a) or early recurrences (Desreumaux et al , 1992; Dubucquoi et al , 1995) of inflammatory diseases, microvascular lesions, and necrosis (Leiferman, 1991; Launay et al , 2000). Release of cationic proteins (ECP and MBP) can also lead to lipid membrane disruption and target cell death (Weller, 1994).…”
