Increased Plasma Transforming Growth Factor‐β1 in Migraine

Abstract: TGF-beta1 in PPP was significantly increased in patients with migraine during headache-free periods. TGF-beta1 may play some role in the development of migraine headache.

“…Interestingly, BDNF is up-regulated in primary sensory neurons by inflammation [10, 26]. Peripheral and cerebrospinal fluid levels of pro-inflammatory cytokines have been found to be elevated in migraineurs [2729]. Cytokine-induced release of pain modulators such as BDNF from trigeminal neurons might underline the interaction of inflammatory and neuronal pathways leading to neurogenic inflammation.…”

“…Furthermore, TGF-␤1 treatment attenuated neuropathic pain (26), and also, TGF-␤ family members were implicated in the modulation of chronic and acute pain perception through the regulation of genes encoding endogenous opioids (27). Also, protein levels of TGF-␤1 are significantly increased in the plasma (25) and cerebrospinal fluid (46) of patients with migraines. To evaluate in vivo the role of TGF-␤1 in nociceptive processes and its involvement in the regulation of Cdk5 activity, we removed TG and DRG from the Tgf-␤1 Ϫ/Ϫ mice and assessed the p35 expression levels and Cdk5 kinase activity in those tissues.…”

“…TGF-␤1 has also been implicated in the pathology of Alzheimer disease (22,23) and its overexpression in astrocytes can lead to excessive deposition of extracellular matrix components in the brain resulting in neurological disease (24). Recent reports indicate that TGF-␤1 may play a role in migraine and neuropathic pain (25)(26)(27). However, the molecular mechanisms underlying its involvement in nociceptive signaling are still far from clear.…”

“…31,32 This study finding of endothelial activation in migraine leads to speculation regarding the nature of the relation. Ictal studies of migraine that have documented increased vWF, 25 endothelin-1, 33 soluble intercellular adhesion molecule, tissue necrosis factor, 34 transforming growth factor-␤1, 35 and matrix metalloproteinase-9 36 suggest that migraine may directly activate the endothelium. An alternative and not mutually exclusive hypothesis is that endothelial dysfunction causes migraine.…”
