“…Mouse mast cells express several different chymases, including mouse mast cell protease 4 (mMCP-4) (Pejler et al 2007), which is the closest functional homologue to the human chymase and mMCP-4 is expressed by connective tissue mast cells in the synovium. Furthermore, the mMCP-4 activity is elevated in the paws and skin of arthritic mice (Kakizoe et al 1999). The mice deficient in mMCP-4 develop significantly less aggressive arthritis compared with the mMCP-4 + / + mice in both the actively and passively induced CIA (Magnusson et al 2009).…”

“…The studies have shown the accumulation of mast cells in the inflamed paws of mice with collagen-induced arthritis (CIA) and imatinib mesylate, inhibitor of multiple tyrosine kinases including kit in mast cells, efficiently suppresses CIA (Kakizoe et al 1999;Paniagua et al 2006). Mast cells have also been shown to accumulate in arthritic lesions of rheumatoid arthritis patients (Maruotti et al 2007).…”

“…The supernatant, which was rich in non-chymase proteases that would interfere with the assay, was discarded and the pellets were homogenized in 5 volumes (w/v) of 10 mM Na-phosphate buffer (pH 7.4) containing 2 M KCl and 0.1% polyoxyethylene octylphenyl ether, stored overnight at 4˚C, and then centrifuged at 15,000 x g for 30 min. The supernatants were used as the extract containing chymase (20). The enzyme activities of chymase were determined using the substrate 0.2 mM N-succinyl-Ala-Ala-Pro-Phe-p-nitroanilide (Sigma) containing 50 μg/ml heparin, 1 mg/ml bovine serum albumin, 1 mg/ml aprotinin, 2 M KCl and 100 mM Tris-HCl, pH 7.6 (21).…”

“…MC tryptase and /or chymase have been shown to destroy the matrix by activating MMPs directly or indirectly (15,29). We have already reported the increase of chymase activity with arthritis in the animal model (19). Furthermore, we have recently observed immunohistochemically that MMPs such as MMP-2, -3 and -9 also showed parallel changes with the severity of arthritis in the animal model (Fig.…”

“…We reported an increase in MC numbers in the arthritic region of the CII-immunized mice (19). Then, we compared CIA of 3 strains of rats among which the severity of arthritic response was different (20).…”
