Indirect somatic embryogenesis from transgenic immature leaf of safflower Carthamus tinctorius (Mohler, Roth, Schmidt & Boudreaux, 1967) (Asterales: Asteraceae)

Abstract: Abstract. Highly efficient protocol is developed for indirect somatic embryogenesis from transgenic immature leaf of safflower Carthamus tinctorius (Mohler, Roth, Schmidt & Boudreaux, 1967) (Asterales: Asteraceae) explants in solid culture. Here two different half strength mediums like MSG and EMM both were tested in addition to inorganic and organic nitrogen sources and also combination of the reference medium. Inorganic nitrogen source shows about 39.5 number of embryos with 37% embryo germination and orga… Show more

“…Explants were carefully dissected (nodal) from 35-day-old in vitro seedlings and cultured in basal nutrient medium Murashige and Skoog (MS), supplemented with B 5 vitamins 3percent ( w / v ) sucrose, 4.52 µM 2,4-Dichlorophenoxy acetic acid (2–4, D), and 2.65 µM α-Naphthalene acetic acid (NAA) (Himedia, Mumbai, India), for callus induction [ 21 , 22 ]. The pH of the medium was adjusted to 5.7 (supplemented with a growth regulator) with 1 N NaOH or 1 N HCl (Himedia, Mumbai, India), before gelling with 0.8percent agar (Himedia, Mumbai, India), and autoclaved at 121 °C for 15 min at 15 lbs pressure.…”

“…Safflower is an erect, herbaceous, highly branched, spiny, thistle-like annual plant that grows from 30 to 150 cm in height (Singh and Nimbkar, 2006;Kumar and Kumari, 2011). Young safflower plants form a rosette and remain in this vegetative state for many weeks, during which leaves and a deep taproot system develop.…”
