Inhibitory Effect of Lithium on p-Aminohippurate Transport in Rat Kidney Cortex in vitro

Abstract: The effect of lithium on p-aminohippurate (PAH) transport was studied using slices and basolateral membrane vesicles prepared from rat kidney cortex. The addition of lithium in concentrations ranging from 0.5 to 5 mM caused a concentration-dependent inhibition of PAH accumulation in the slices. Lithium inhibited PAH accumulation in the slices, not only during the rapid uptake period (after 10 min) but also during the approach to equilibrium (after 30 min). The effect of lithium (2 mM) in the slices was irrever… Show more

“…), and their kidneys were rapidly removed, decapsulated and placed in ice-cold isotonic saline medium. Renal cortical slices were prepared freehand using a razor blade on an ice-cold petri dish and were placed in ice-cold medium containing 134 mM NaCl, 5.9 mM KCl, 1.5 mM CaCl2, 1.2 mM MgCl2, 11.5 mM glucose and 5.8 mM N-2-hydroxyethylpiperazine-N'-2-ethanesulfonic acid (HEPES), titrated with NaOH at pH 7.4 as already described previously (20). The slices were transferred to flasks containing the same medium.…”

“…PAH accumu lation was calculated as the ratio of concen tration of PAH in the intracellular fluid (S) to that in the medium (M), the S/M ratio. The experimental methods of the slice-study were essentially the same as those already described (9).…”

“…The 30-min incubation time for study of PAR accumulation in rat renal cortical slices is sufficient for PAM to reach equilibrium [13,21]. We found that prior stevioside treatment in vivo significantly reduced PAR accumulation in the renal cortical slices of rats.…”
