Inhibitory spectrum of alpha 2-plasmin inhibitor.

Abstract: ABSTRACTarPlasmin inhibitor (a2PI) has been recently characterized as a fast-reacting inhibitor of plasmin in human plasma and appears to play an important role in the regulation of fibrinolysis in vivo. We have studied the effect of purified a2PI upon various proteases participating in human blood coagulation and kinin generation. At physiological concentration (50 ;sg/ml), a2PI inhibited the clot-promoting and prekallikrein-activating activity of Hageman factor fragments, the amidolytic, kininogenase, and cl… Show more

“…Although a2PI is also known to inhibit some activated clotting factors (Xla or thrombin) in purified systems (33,34), there was no significant abnormality in blood coagulation tests in this patient's plasma. The abnormally increased fibrinolytic activity was solely ascribed to the severe deficiency of a2PI, because the addition of purified a2PI to the patient's blood completely corrected its abnormality.…”

“…4) (I). The values for k' were tabulated for a1-protease inhibitor, antithrombin III, and Cl-inhibitor from this study, and for a2-plasmin inhibitor from the report of Saito et al (7). From these data we predicted that al-protease inhibitor would be responsible for 68% of the inhibitory activity in plasma against Factor XIa.…”

“…The clot formed was suspended in normal plasma and incubated at 370C. At 2 (-), 6 (0), and 24 (A) h of incubation, release of radiolabel from the clot into the suspending plasma milieu was measured and expressed as a percentage of the total radioactivity. The concentrations of a2PI in the clot at the time of clotting are given on the abscissa.…”
