Insertions in the gG Gene of Pseudorabies Virus Reduce Expression of the Upstream Us3 Protein and Inhibit Cell-to-Cell Spread of Virus Infection

Abstract: The alphaherpesvirus Us4 gene encodes glycoprotein G (gG), which is conserved in most viruses of the alphaherpesvirus subfamily. In the swine pathogen pseudorabies virus (PRV), mutant viruses with internal deletions and insertions in the gG gene have shown no discernible phenotypes. We report that insertions in the gG locus of the attenuated PRV strain Bartha show reduced virulence in vivo and are defective in their ability to spread from cell to cell in a cell-type-specific manner. Similar insertions in the g… Show more

“…Although appreciable numbers of double-infected neurons are observed within the medulla and pons at 120 h after injection (Kerman et al, 2003). A single replication cycle of the virus is 10 -12 h (Demmin et al, 2001). We therefore decided to extend the survival times by 12 h to 132-136 and 144 h. In preliminary experiments, we determined that there is maximal labeling in the forebrain and midbrain at 144 h of survival, and we thus used that as the longest survival time point.…”

“…Third, since US3null PRV displays a small plaque phenotype (Demmin et al, 2001) (Fig. 3c), we checked whether inhibiting apoptosis increased the plaque size of US3null PRV or, as a control, WT PRV.…”

“…28 Not surprisingly, reduction in plaque size is a property associated with Us3 null viruses. 7 An example of the notable reduction in cell-to-cell spread of PRV in the absence of Us3 is shown in Figure 1B. Very recently, Van den Broeke and co-workers demonstrated that PRV Us3 interacts with and phosphorylates the p21-activated kinases, Pak1 and Pak2, and that Pak1 is specifically required for PRV Us3 induced FP formation.…”

“…7 These studies were followed by the observation that transient expression of PRV Us3 in cultured cells resulted in the formation of long filamentous processes (FPs) that contained both actin and microtubules. 28,30 An example of PRV Us3-induced FP formation is shown in Figure 1A.…”
