Insight into the ERVK Integrase – Propensity for DNA Damage

Abstract: Retroviruses create permanently integrated proviruses that exist in the host genome. Retroviral genomes encode for functionally conserved gag, pro, pol, and env regions, as well as integrase (IN), which is required for retroviral integration. IN mediates viral genome insertion through 3′ end processing of the viral DNA and the strand transfer reaction. This process requires the formation of a pre-integration complex, comprised of IN, viral DNA, and cellular proteins. Viral insertion causes DNA damage, leading … Show more

“…Based on homology modeling, we predict that the ERVK IN enzyme contains all the essential motifs and domain structures for retroviral IN function [ 22 ]. A recombinant ERVK-10 integrase enzyme also confirms that it has the potential for strand-transfer activity [ 23 ]. A remaining question is how ERVK IN interacts with cellular proteins and pathways, as has been shown for many other retroviral integrases [ 19 , 24 , 25 , 26 ].…”

“…Those HML-2 RT enzymes were strictly dependent on Mg2 +, and did not show polymerase activity with other divalent cations (Berkhout et al 1999). The integrase of HML-2 (HERV-K10) has also been cloned; it showed both terminal cleavage and strand transfer in the presence of Mn2 + of the HML-2 LTR substrate (Kitamura et al 1996; Bray et al 2016). In the human genome (GRCh38) only 8 HML-2 proviruses have intact integrase motifs (Bray et al 2016).…”

“…Enzymatic activity of retroviral integrase proteins can lead to significant DNA damage accumulation over time (49). Given a loss of DNA repair function in SBMA, this may render cells more vulnerable to DNA damaging insults like ERVK integrase activity (21) (unpublished data). Therefore, our observation of mature ERVK integrase protein expression in PBMC from patients with AR polyQ repeats is potentially pathologically relevant to the underlying multi-system disease processes that occur in SBMA.…”
