Instrumentation for Vibrational Circular Dichroism Spectroscopy: Method Comparison and Newer Developments

Abstract: Vibrational circular dichroism (VCD) is a widely used standard method for determination of absolute stereochemistry, and somewhat less so for biomolecule characterization and following dynamic processes. Over the last few decades, different VCD instrument designs have developed for various purposes, and reliable commercial instrumentation is now available. This review will briefly survey historical and currently used instrument designs and describe some aspects of more recently reported developments. An import… Show more

“…Vibrational circular dichroism (VCD) may be considered for directly unveiling the chirality of chitin nanocrystals in aqueous medium. This technique has been proven to work in solution or gel states 49 and has been useful to identify peptide and protein fiber chiral features and to distinguish related variation in conformations. 50 Noting that interactions with water significantly alter the spectra and that a sufficiently high concentration is needed for VCD analysis, its application to chitin nanocrystals needs to be evaluated.…”

“…A technique that has surely not yet developed all its potential is vibrational circular dichroism (VCD). , This is a technique offering interesting possibilities as the theoretical modeling of the experimental spectra is simpler than for UV–vis spectra. New instrumentation is being developed to increase the instrument sensitivity for weak dichroic signals, but it is mostly optimized for specific problems, for example, for chiral studies on biopolymers …”

“…Consequently, one can scan relatively quickly over the band and coadd multiple spectra to improve S/N, without spending measurement time or acquiring noise from spectral regions of no interest. These instrument designs have been reviewed and compared in some detail in the past. ,,,,,,,,,,…”

“…In particular, conference proceedings and meeting abstracts are not referenced. However, there are a great many previous reviews in this field, which tend toward a more general nature and provide added background (refs , , , , , , , − , − , ). Those that emphasize biopolymer studies with VCD or ROA will be referenced as appropriate to enable the reader to access older work in more detail, leaving this review to emphasize more focused developments.…”
