Insulin-like growth factor 2 mRNA binding protein 2 promotes aerobic glycolysis and cell proliferation in pancreatic ductal adenocarcinoma via stabilizing GLUT1 mRNA

Abstract: Insulin-like growth factor 2 mRNA binding protein 2 (IGF2BP2) is a member of the IGF2BP protein family consisting of IGF2BP1~3 with the capacity of binding to many transcripts and regulating RNA stability, localization, and translation. In this study, we discovered that expression of IGF2BP2 was upregulated and led to a poor prognosis in pancreatic ductal adenocarcinoma (PDAC). IGF2BP2 protein was gradually elevated from normal pancreas, pancreatic intraepithelial neoplasia to PDAC in an LSL-KrasG12D/+;LSL-Trp… Show more

“…For this study, we focused on the PoRs (Figure 3A), since these represent factors that may promote dedifferentiation and diseases progression and may thus present therapeutic targets eligible for treatment by inhibition, as proposed for LIN28B [29] and other RBPs. Among the 44 PoRs were proteins previously implicated in PDAC progression, e.g., IGF2BP1-3 [13][14][15][16][17], IFIT3 [30], EXO1 [21], or PTBP3 [23]. However, there were also proteins that, to the best of our knowledge, have not yet been associated with oncogenic potential in pancreatic cancer, e.g., OAS proteins, RBM34 or DQX1.…”

“…Despite the lack of systematic investigation of RBPs in PDAC, ample evidence suggests substantial impact of some RBPs in this malignancy. For example, all three members of the insulin-like growth factor 2 mRNA binding protein family (IGF2BPs) have recently been described as being upregulated and associated with a poor prognosis in pancreatic cancer [13][14][15][16][17]. Furthermore, inactivation of the histone deacetylase SIRT6 was reported to accelerate PDAC progression in mouse models due to upregulation of LIN28B [18].…”

“…In recent years, over-expression of IGF2BP2 in multiple human cancers has been associated with poorer prognosis of the disease. For instance, over-expression of IGF2BP2 confers shorter survival and poor prognosis of patients with acute myelocytic leukemia (AML) [65], breast cancer [14], esophageal carcinoma [16], low-grade gliomas [17], hepatocellular carcinoma (HCC) [66], head and neck squamous cell carcinoma (HNSCC) [67], pancreatic ductal adenocarcinoma (PDAC) [20,[68][69][70] and gallbladder carcinoma (GBC) [71]. Herein, we summarize the specific roles of IGF2BP2 in multiple cancers and provide a comprehensive view of IGF2BP2 ( Fig.…”

“…IGF2BP2 is also over-expressed in PDAC [68,69]. Meanwhile, Glucose transporter 1 (GLUT1) is an integral membrane protein consisting of 12 transmembrane helices and an intracellular domain which promotes aerobic glycolysis and proliferation of cancer cells [75][76][77].…”

“…CNP was correlated with survival and could be used as a novel and potentially useful clinical prognosis biomarker in glioblastoma multiforme [39]. The expression of IGF2BP2 was up-regulated and led to a poor prognosis in pancreatic ductal adenocarcinoma [40] and acute myelocytic leukemia [41]. PABPC1L was emerging as a promising prognostic factor for colon cancer [42].…”
