Integrated Metabolomic and Transcriptomic Analysis Reveals the Effect of Artificial Shading on Reducing the Bitter Taste of Bamboo Shoots

Abstract: Bamboo shoot is a delicious and nutritious forest vegetable. It has been found that bamboo shoots collected from low-light environments have a less bitter taste. The molecular mechanism of light in the regulation of bitter substance accumulation in bamboo shoots is still unclear. In this study, we applied a shading treatment to Pleioblastus amarus bamboo shoots in the preharvesting period. The reduction in the bitterness intensity was confirmed by a sensory test. An integrated metabolomic and transcriptomic an… Show more

“…Coupled with mass spectroscopy (MS), LC–MS is ideally suited for detecting and quantifying multi-components in complex plant extracts [ 52 ]. Based on our previous work on the chemical constituents of bamboo plants [ 38 , 53 , 54 , 55 ], 21 phenolic compounds were selected as the extraction targets in this research. The molecular structures of the target compounds are shown in Figure S3 .…”

“…The RNA was extracted from leaf samples using a modified CTAB method ( Kanani et al 2019 ). Reverse transcription (RT) was conducted with the Monad 1st Strand cDNA Synthesis Kit, and the qRT-PCR analysis was achieved using Tb Green® Premix Ex Taq™ II (Takara, Beijing, China) as previously described ( Huang et al 2022 , 2023 ). All samples had three biological and technical replicates.…”

“…The UPLC-MS/MS analysis of anthocyanin compounds was based primarily on the detailed standard procedure described previously [44]. To determine the substrate specificity for PgF3 5 H, nine flavonoids (Table 1), which can be directly catalyzed or synthesized by F3 5 H [12,45,46], were chosen as standards for further testing.…”
