Integrin ?1 localization in murine central and peripheral nervous system

Abstract: The distribution of neurons expressing integrin alpha1 subunit protein (INTalpha1) was examined in adult mouse tissues of not only the central nervous system, but also the sympathetic ganglia, and the adrenal gland by immunohistochemistry and immunoelectron microscopy. INTalpha1-positive neurons were observed in most tissues examined, and most of them were found to coexpress tyrosine hydroxylase (TH) except for Purkinje cells and hippocampal neurons. Expression of INTalpha1 was also observed in the malposition… Show more

“…42,43 The higher bands (␣1, 230 kDa; ␣2, 200 kDa) may represent posttranslational modifications, with the 230-kDa product described in mouse tissue. 42 To confirm further the presence of integrin protein, scleral fibroblasts were colabeled with the ␣2␤1 integrin antibody and an antibody to vinculin, a structural protein present in focal adhesions. 44 Figure 3 shows that ␣2␤1 integrin and vinculin (Figs.…”

“…Although only a 30 -40% reduction was observed, these results are significant, and even a bit remarkable, given the wide number of adhesive interactions that might contribute to axon extension. Sympathetic neurons express a number of integrins, including the ␣ 1 and ␣ 3 subunits, and many integrin ligands and cell adhesion molecules are present in pathways followed by sympathetic axons (Reichardt et al, 1990;DeFreitas et al, 1995;Murase and Hayashi, 1998).…”

“…For example, alpha-4 mRNA was detected only in discrete regions of the limbic forebrain. Integrin protein immunoreactivity has also been detected in brain for the alpha-1, alpha-3, alpha-5, alpha-8, alpha-v, beta-1, beta-3, beta-5 and beta-8 subunits (45, 124,[127][128][129][130][131][132][133][134][135]. In some cases, integrin subunit protein staining is seen in regions where the mRNA is not detected ( e.g., alpha-8, (132)), indicating that distribution may be wider than revealed by in situ hybridization.…”
