Interaction of a Potential Anticancer Agent Hypericin and its Model Compound Emodin with DNA and Bovine Serum Albumin

Abstract: Anticancer agent hypericin and its derivative emodin interact with DNA with medium strength and are probably incorporated into the groove of DNA by hydrogen bonds. Bovine serum albumin can serve as a transport protein for hypericin since the binding force between both molecules is adequate.

“…This interaction is characterized by binding constant 8.1 × 10 4 L/mol. These ligands are incorporated into the DNA groove with hydrophobic or hydrogen bounds [204]. An intercalating mode of interaction with DNA has been confirmed for quinizarin and danthron, which is very clearly supported by atomic force microscopy measurements.…”

“…As a representative of highly hydrophobic PSs, hypericin binds to DNA by means of a large groove [203,204]. A binding constant, whose value was also determined, is 4.0 × 10 4 L/mol [204]. Emodin, with mild hydrophobic properties, interacts with DNA by binding into a small groove.…”

“…A plethora of studies have been conducted to investigate the interaction of various antitumor, anticancer, antiviral, and anti‐inflammatory drugs with DNA as well as serum albumin (bovine or human) protein [30–41]. Moreover, the interaction between three antimycobacterial drugs (dapsone, clofazimine, and rifampicin) and bovine or human serum albumin (HSA or BSA) have also been evidenced by different groups of scientists [42–47].…”

[25] Common serum albumins include human serum albumin and bovine serum albumin, which are involved in various transportation and regulation of the body system. [26] The interaction of a compound with BSA can enhance its solubility and dispersibility during blood transport.…”
