Interaction of the cell-binding domain of fibronectin with VLA-5 integrin induces monokine production in cultured human monocytes

Abstract: SUMMARYThe effect of fibronectin on IL-1a, IL-1j3, tumour necrosis factor-alpha (TNF-a), and IL-6 production was investigated with cultured monocytes isolated from human peripheral blood. Monokine concentrations were determined by both ELISA and bioassay. Fibronectin markedly stimulated the secretion of IL-1 a, IL-1,6, TNF-a and IL-6 from cultured monocytes in a dosedependent manner, with the maximal effect apparent within 24 h. Northern blot analysis revealed a marked increase in the abundance ofmRNA specific… Show more

“…rsCD40L Induced IL-8 Gene Expression in U937 Cells-The interaction of monocytes with fibronectin leads to inflammatory cytokines expression (30,32), cellular responses that could also be induced in these cells by sCD40L-triggered signaling. To assess the expression of biological mediators, we analyzed IL-8 gene expression in U937 cells stimulated with 100 ng of rsCD40L for 15 min to 4 h. In U937 cells, rsCD40L induced a weak but significant IL-8 gene expression (Fig.…”

“…However, the physiological role of plasma FN is still unknown. We have previously investigated the effect of the soluble form of FN on the production of IL‐1 α , IL‐1 β , IL‐6 and TNF‐ α by cultured human monocytes from normal individuals [10]. FN markedly stimulated the secretion of IL‐1 α , IL‐1 β , IL‐6 and TNF‐ α .…”

“…In cultured human monocytes, FN‐coated substrate enhanced both monocyte adhesion and cytokine production. We have previously shown that FN in culture medium activates monocytes through the VLA‐5 receptor, inducing the production of proinflammatory cytokines such as IL‐1, IL‐6 and tumour necrosis factor‐alpha (TNF‐ α ) by these activated cultured human monocytes [10]. Many studies have shown that these same exogenous cytokines are also able to stimulate cultured human monocytes [1–3].…”

“…Furthermore, exosomes from several independent mouse tumor cell lines trigger MDSC suppressive function in an HSP72/TLR2 dependent manner 63 . At the level of macrophages, exosome-associated fibronectin induces IL-1β production 64 and microvesicles released by late stage mouse melanoma cells were found to exert a dose-dependent suppression of MHC Class II expression in these cells 65 . Recently, it has been proposed that miRNAs from tumor exosomes may also play a role in mobilizing myeloid cells to pre-metastatic niches 3 .…”
