Interactions among Peroxisome Proliferator Activated Receptor-γ, Insulin Signaling Pathways, and Steroidogenic Acute Regulatory Protein in Human Ovarian Cells

Abstract: Insulin and TZDs independently stimulate expression of PPAR-gamma, insulin receptor, IRS-1, and StAR protein in human ovarian cells. Thus, PPAR-gamma, insulin receptor with its signaling pathways, and StAR protein constitute a novel human ovarian regulatory system with complex interactions among its components.

“…However, inconsistency in their expression and activity has been often demonstrated by researchers. For instance, a stimulatory effect of PPARg activation on steroidogenesis can be mediated by up-regulation of steroidogenic acute regulatory (StAR) protein in human and macaque granulosa cells [44,45] or by 3b-HSD in porcine ovarian follicles [36]. An opposite effect of PPARg ligands on 3b-HSD expression and P 4 release was observed in porcine granulosa cells [37].…”

“…Such a relationship has been observed in ovarian cells in different species, but the results are contradictory. For instance, PPARγ ligands enhanced P 4 release by porcine theca cells (Schoppee et al 2002) and mixed human ovarian (stroma, theca, and granulosa) cells (Seto-Young et al 2007), but inhibited hormone secretion by porcine and hu- Table 2. “…The procedure for the collection and incubation of endometrial tissue was described previously (Bogacka et al 2013b (Lovekamp et al 2001;Schoppee et al 2002;Seto-Young et al 2007). The tested compounds were dissolved in a total volume of 20 μl dimethyl sulfoxide (DMSO) (Sigma-Aldrich).…”
