BackgroundPrevious studies have suggested that IL4, IL13, and IL4R are associated with serum IgE levels and allergies, and common variants of these genes may alter cancer risk. To clarify these associations, we conducted a meta-analysis to investigate the associations of IL4, IL13, and IL4R polymorphisms with gastrointestinal cancer risk.MethodsWe used 27 eligible case–control studies describing the associations of six polymorphisms of IL4, IL13, and IL4R with gastrointestinal cancer risk to calculate summary odds ratios (ORs) and 95% confidence intervals (CIs) using five different genetic models. The Q-statistic and I2 statistic were calculated to examine heterogeneity.ResultsThe IL4 rs2070874 T allele seems to be associated with an increased risk of gastrointestinal cancer (OR 1.11; 95% CI, 1.00–1.24 for T allele vs. C allele). This association was significant in studies conducted outside of Asia (OR 1.28; 95% CI, 1.03–1.58 for T allele vs. C allele) and in studies investigating the association with gastric cancer (OR 1.17; 95% CI, 1.03–1.34 for T allele vs. C allele). However, the IL4R rs1801275 heterozygote seems to be associated with a reduced risk of gastrointestinal cancer (OR 0.79; 95% CI, 0.65–0.96 for AG vs. AA). Other polymorphisms did not show any significant associations with gastrointestinal cancer risk in any of the genetic models and subgroup analyses.ConclusionsOur results suggest that certain polymorphisms of IL4 and IL4R may affect susceptibility to gastrointestinal cancer. However, further studies are required to confirm these findings.

AIMTo study the genetic association and epistatic interaction of the interleukin (IL)-10 and IL-10/STAT3 pathways in pediatric inflammatory bowel disease (IBD).METHODSA total of 159 pediatric inflammatory IBD patients (Crohn’s disease, n = 136; ulcerative colitis, n = 23) and 129 matched controls were studied for genetic association of selected single nucleotide polymorphisms (SNPs) of the IL-10 gene and the genes IL10RA, IL10RB, STAT3, and HO1, from the IL-10/STAT3 signaling pathway. As interactions between SNPs from different loci may significantly affect the associated risk for disease, additive (a) and dominant (d) modeling of SNP interactions was also performed to examine high-order epistasis between combinations of the individual SNPs.RESULTSThe results showed that IL-10 rs304496 was associated with pediatric IBD (P = 0.022), but no association was found for two other IL-10 SNPs, rs1800872 and rs2034498, or for SNPs in genes IL10RA, IL10RB, STAT3, and HO1. However, analysis of epistatic interaction among these genes showed significant interactions: (1) between two IL-10 SNPs rs1800872 and rs3024496 (additive-additive P = 0.00015, Bonferroni P value (Bp) = 0.003); (2) between IL-10RB rs2834167 and HO1 rs2071746 (dominant-additive, P = 0.0018, Bp = 0.039); and (3) among IL-10 rs1800872, IL10RB rs2834167, and HO1 rs2071746 (additive-dominant-additive, P = 0.00015, Bp = 0.005), as well as weak interactions among IL-10 rs1800872, IL-10 rs3024496, and IL-10RA (additive-additive-additive, P = 0.003; Bp = 0.099), and among IL10RA, IL10RB, and HO1 genes (additive-dominant-additive, P = 0.008, Bp = 0.287).CONCLUSIONThese results indicate that both the IL-10 gene itself, and through epistatic interaction with genes within the IL-10/STAT3 signaling pathway, contribute to the risk of pediatric IBD.

Gastric cardiac adenocarcinoma (GCA) is one of common malignant tumors in the world. Multiple genes that play critical roles in inflammatory pathways probably are associated with GCA risk. We conducted a hospital-based case-control study to evaluate the genetic effects of functional single nucleotide polymorphisms (SNPs): interleukin 9 (IL9) rs31563 C > T, IL9 rs31564 G > T, IL10 rs1800872 T > G, IL12A rs2243115 T > G, IL12B rs3212227 T > G, and IL13 rs1800925 C > T on the development of GCA. Two hundred and forty-three GCA cases and 476 controls were recruited. Their genotypes were determined using a custom-by-design 48-Plex SNPscan kit. IL12B rs3212227 T > G polymorphism was associated with the increased risk of GCA. However, there was no significant association between the other five SNPs and GCA risk. Stratified analyses indicated that the risk of GCA associated with the IL12B rs3212227 T > G polymorphism was evident among female patients and patients who never smoked or consumed alcoholic drinks. These findings indicated that functional polymorphism IL12B rs3212227 T > G might correlate with GCA risk. However, our results were obtained with a limited sample size; the power of our analysis was low. Larger studies are required to confirm the current findings.
