Interleukin 37 promotes angiogenesis through TGF-β signaling

Abstract: IL-37 is a novel pro-angiogenic cytokine that potently promotes endothelial cell activation and pathological angiogenesis in our previous study, but the mechanisms behind the pro-angiogenic effect of IL-37 are less well understood. Extending our observations, we found that TGF-β interacts with IL-37, and potently enhances the binding affinity of IL-37 to the ALK1 receptor complex, thus allowing IL-37 to signal through ALK1 to activate pro-angiogenic responses. We further show that TGF-β and ALK1 are required i… Show more

“…TGF-β is a pivotal regulator both in pathophysiological angiogenesis and tissue remodeling [ 31 33 ]. In microvascular endothelial cell lines, TGF-β1 can promote angiogenesis by regulating thrombospondin-4 (TSP-4) [ 34 ], and IL-37 exerts its pro-angiogenic responses through the TGF-β-ALK1 signaling pathway [ 35 ]. A previous study showed that the inhibition of PI3K signaling might be a promising therapeutic target for thrombotic diseases [ 36 ].…”

“…These results demonstrated that the antitumor role of IL‐37 in NSCLC development could be through inhibiting tumor angiogenesis. However, some studies have suggested IL‐37 as a proangiogenic factor, which needs further investigation.…”

“…Moreover, this proangiogenic effect was not influenced when IL‐1R8 or IL‐18Rα were neutralized, indicating that other unknown receptors are involved in this process. Two years later, the same research group identified this unknown receptor as ALK1 receptor complex . They found that TGF‐β may act as the bridging molecular that mediates IL‐37 binding to the TGF‐β receptor complex, thus promoting the angiogenesis process.…”
