Interleukin 39: a new member of interleukin 12 family

Abstract: interleukin (iL)-12 family member is a heterodimer glycoprotein, composed of two covalently linked subunits, α and β chains. The α subunit consists of iL-23p19, iL-27p28, and iL-12p35, and the β subunit includes iL-12p40 and epstein-Barr virus-induced gene (ebi3). iL-39 is a new heterodimeric iL-12 family member composed of iL-23p19 and ebi3 subunits. iL-39 is secreted by lipopolysaccharide-stimulated B cells. other immune cells, such as dendritic cells and macrophages, express iL-39 mRna. in lupus-like mice, … Show more

“…IL-23/IL-39 inhibitors are a class of biologics used to treat moderate to severe plaque psoriasis. IL-39, a heterodimer composed of IL-23p19 and Epstein-Barr virus-induced gene 3, is a new member of the IL-12 family and is targeted by p19 inhibitors [ 96 ]. Specifically, in contrast to IL-12/23 inhibitors, IL-23/39 inhibitors inhibit IL-23 without inhibiting IL-12.…”

“…Interleukin-35 (IL-35) is an IL-12 family cytokine (12), with other members of these family consisting of heterodimeric combinations of a (p40 and EBI3) and b (p19, p28 and p35) chain subunits, including IL-12 (p35 and p40), IL-23 (p19 and p40), IL-27 (p28 and EBI3), IL-35 (p35 and EBI3), and the newly discovered member IL-39 (IL-23p19 and EBI3) (13,14). Unlike other IL-12 family members, IL-35 has been shown to exhibit immunosuppressive activity (15).…”

“…This also applies to their molecular weight (11.098–141.752 kDa), isoelectric point (from 4.41 for IL-18 to 10.5 for IL-11) and the contents of the hydrophilic and hydrophobic amino acids in the composition of a molecule ( Table 1 ). Most of the interleukins are monomeric compounds, with the exception of IL-35, which is composed of the subunits IL-12 α and IL-27 β , and IL-39, composed of IL-23p19 and Ebi3 (Epstein–Barr virus-induced gene) subunits [ 45 ]. Detailed characterization of the interleukins based on the aforementioned properties, as well as the analysis of the secondary structure, is presented in Table 1 .…”
