Interleukin-8 can mediate acute-phase protein production by isolated human hepatocytes

Abstract: During the course of studies designed to identify the role of cytokines in the reprioritization of hepatic protein synthesis associated with cachexia we detected a hepatocyte-stimulating moiety in the supernatants of pancreatic cancer cells that was unrelated to interleukin (IL)-6. This study identifies that moiety as IL-8 and investigates the role of IL-8 in the induction of acute-phase protein production. The human pancreatic cancer cell line MIA PaCa-2 produced >1 ng/ml of IL-8 per 24 h, and supernatants… Show more

“…Thus these cytokines could be implicated in a specific response to sole ulcers. In turn, IL-1β can increase expression of CXCL8 mRNA in leukocytes (Fujishima et al, 1993), and both IL-1β and CXCL8 are implicated in activating haptoglobin secretion (Godson et al, 1995;Wigmore et al, 1997). This is in agreement with the tendency for higher serum haptoglobin, CXCL8 mRNA, and haptoglobin mRNA expression in the lame cows in this study.…”

“…20 Human hepatoma cell line HepG2 is able to produce CRP Ϸ4-fold compared with control in conditions similar to those used in our experiments. 21 Another hepatoma cell line Hep3B stimulated with conditioned medium (generated by stimulating peripheral blood mononuclear cell with LPS at a dose of 1 g/mL for 24 hours) showed an Ϸ50-fold increase in CRP mRNA compared with unstimulated cells.…”

“…IL-8 has not been isolated from rats, and CINC-1 is considered to be the rat counterpart of human IL-8 [25,26]. IL-8 was confirmed to mediate the production of acute-phase proteins in isolated human hepatocytes [28]. The serum levels of α2M after injection of the IL-6-or CINC-1-rich fractions increased; however, no significant differences were observed when compared with controls.…”
