Interleukins 4 and 21 Protect Anti-IgM Induced Cell Death in Ramos B Cells: Implication for Autoimmune Diseases

Abstract: Interleukins 4 (IL-4) and 21 (IL-21) belong to the common gamma chain cytokine family which are highly involved in the progression of autoimmune diseases. While IL-4 is well known to be involved in the suppression of apoptosis of autoreactive B cells, the role played by IL-21 remains unclear. In the current study, we activated the human Burkitt’s lymphoma Ramos B cells with anti-IgM to mimic B cell hyperactivation observed in patients of autoimmune diseases. Consistent with other reported findings, anti-IgM le… Show more

“…IL4 levels were elevated in all four groups of patients with autoimmune diseases (rheumatoid arthritis, Sjögren's syndrome, systemic lupus erythematosus and ulcerative colitis) (Supplemental Fig. S6), consistent with the role of this protein in B-cell hyperactivity, which mediates a broad spectrum of autoimmune diseases 22 . IL17A and IL17F are cytokines secreted by T helper 17 cells that play crucial roles in the was not certified by peer review) is the author/funder.…”

“…Discontinuous fragmentation of nuclear DNA during apoptosis is revealed by discrete “Sub-G1” peaks on DNA content histograms [ 94 ]. Cells accumulated in the sub-G0/G1 phase are classified as apoptotic cells with DNA fragmentation and mitochondrial dysfunction [ 95 ].…”

“…9). IL4, IL17 and IL33 all have well established roles in autoimmune diseases [21][22][23] . These results thus indicate that the higher sensitivity of NULISAseq can identify more clinically important but low-abundance biomarkers.…”
