Internalization and Down-regulation of Human Muscarinic Acetylcholine Receptor m2 Subtypes

Abstract: ؊4 M carbamylcholine for 16 h. However, the rate of down-regulation was lower compared with wild type receptors (t1 ⁄2 ‫؍‬ 9.9 versus 2.3 h). These results indicate that rapid internalization of hm2 receptors is facilitated by their phosphorylation with GRK2 and does not occur in the absence of the third intracellular loop, but downregulation of hm2 receptors may occur through both GRK2-facilitating pathway and third intracellular loopindependent pathways.Loss of the cell's response to agonist acting at G prot… Show more

“…Interestingly, the observation that a mutant form of the m2-muscarinic receptor that does not undergo agonist-promoted endocytosis can be down-regulated (59) suggests that endocytosis-independent down-regulation can also be observed for other receptors.…”

“…Phosphorylated receptors then interact with cytoplasmic proteins termed ␤-arrestins, which interfere with receptor-G protein interaction, favoring receptor endocytosis, thus inhibiting the signal (49,50). Generally muscarinic receptor-linked PLD activity is inhibited rapidly within 2 min, but the completion of muscarinic receptor phosphorylation and internalization events are required for a longer time (51,52). Therefore, muscarinic receptor-linked PLD activity might be inhibited by another mechanism.…”

“…The adenylyl cyclase-coupled m2-muscarinic receptor was one of the first receptors to be shown to be an in vitro substrate for the GRKs (31). Phosphorylation at sites on the third intracellular loop is thought to mediate m2-muscarinic receptor internalization (6,32). m3-muscarinic receptors contained in urea-treated membranes have previously been shown to be phosphorylated by purified GRK-2 and -3 but not GRK-5 and -6 (11).…”

“…Originally associated with receptor desensitization (1,2), GPCR phosphorylation has now been implicated in a number of processes including receptor internalization (3)(4)(5)(6) and as a molecular switch that determines coupling to specific signaling pathways (7,8). The receptor-specific kinases involved are generally considered to belong to the G-protein-coupled receptor kinase (GRK) family which are characterized by their sequence homology to rhodopsin kinase (GRK-1) and that include the extensively studied ␤-adrenergic receptor kinases 1 and 2 (GRK-2 and -3, respectively) (2,9).…”
