Intestinal microbiota link lymphopenia to murine autoimmunity via PD-1+CXCR5−/dim B-helper T cell induction

Abstract: T cell lymphopenia results in peripheral homeostatic expansion to maintain the T cell immune system, which is termed lymphopenia-induced proliferation (LIP). LIP is a potential risk for expanding autoreactive clones to become pathogenic in human and murine autoimmune diseases. However, the ontogeny of T cells that induce autoantibody production by autoreactive B cells in LIP remains unclear. Transfer of CD4+CD25− conventional T (Tc) cells into T-cell-deficient athymic nude mice has been previously reported as … Show more

“…S3, available at Rheumatology Advances in Practice online). Given that it has been reported that PD-1 + CXCR5 − /dim CD4 + T cells and Tph cells also have a functional property as B-cell helpers [5, 13], we performed functional analysis regarding the promotion of B-cell responses. However, we failed to observe significant levels of expression of IL4 and IL21 transcripts in Tph-like cells from healthy volunteers and from patients with IgG4-RD compared with those in Tfh cells (supplementary Fig.…”

“…In multiple sclerosis, a decrease in Butyricimonas content resulted in SCFAs reduction, which attenuated the anti-inflammatory effects of intestinal Treg cells [34]. The study of Eri T et al also showed that the reduction of Odoribacter and Alistipes induced by long-term use of antibodies may damage intestinal barrier function and induce intestinal sepsis [35]. Based on these previous reports and our experimental results, we found that abnormal microbiome composition mediated by Drp1 could reduce the colon SCFAs level, resulting in the destruction of tight junction in IECs and the increase of intestinal permeability after shock.…”

“…43 In turn, manipulation of intestinal microbiota in mice has been shown to directly modulate lymphopenia-induced autoimmunity. 44 Lymphopenia may be a feature of pre-diagnostic PD, and reflect early pathological changes in peripheral tissues.…”
