Intranasal immunization with a recombinant outer membrane protein H based Haemorrhagic septicemia vaccine in dairy calves

Abstract: Haemorrhagic septicemia (HS) is a contagious disease in cattle with high morbidity and mortality rates. HS vaccine in Thailand is an oil-adjuvant formulation, and is difficult to administer. The present study aimed to formulate and evaluate the protection in dairy calves conferred by immunization with an in-house intranasal HS vaccine. The intranasal vaccine was formulated in a total volume of 500 µl containing either 50 or 100 µg of the recombinant outer membrane protein H (rOmpH) of Pasteurella multocida str… Show more

“…Intranasal treatment with P multocida outer membrane protein H induced both serum IgG and secretory IgA levels that protected calves from experimental challenge with P multocida. 49 Although studies have yet to be conducted in cattle, a recombinant outer membrane lipoprotein B from P multocida serotype A strain was shown to induce serum Ab in mice with significant bacterial killing activity. 50 Regarding H somni immunity, antibodies to a 40-kDa outer membrane protein (OMP) have been found protective, whereas those to a 78-kDa OMP are not.…”

“…Various formulations of HS vaccines are available to treat animals, including inactivated vaccines, live vaccines, purified capsular extract vaccines, and combined vaccines [2]. e parenteral administration of HS is an oiladjuvant formulation, but it is inconvenient for practical use and may induce stress in animals [10]. Although several conventional vaccine formulations are commercially available, the quest for suitable broadly protective HS vaccines with long-lasting immunity is on the upsurge [4].…”

“…Up to date, a variety of modern vaccines, including recombinant vaccines, have been developed as veterinary vaccine candidates for HS prevention [11]. e outer membrane protein H (OmpH) is a surface antigen of P. multocida, recognized as an immunodominant porin and potential target to be a vaccine candidate for pasteurellosis in domestic animals [10,[12][13][14][15]. Concerning the buffaloes and cattle, the 37 kDa OmpH of P. multocida serotype B:2 has been identified and suggested as a highly antigenic protein [16][17][18][19].…”
