Abstract. Barley stripe rust (BSR), caused by Puccinia striiformis f. sp. hordei, has been a major disease problem to the barley industry worldwide that has not, to date, been detected in Australia. This paper describes the mapping of stripe rust resistance genes in Tallon/Kaputar (TK) and Arapiles/Franklin (AF) populations. The paper also reports on the usefulness of markers associated with alternative sources of resistance previously identified in the varieties Orca and Shyri in the USA. Stripe rust screening was conducted at the adult plant stage in Toluca, Mexico, for 2 years. Two major quantitative trait loci (QTLs) were found on chromosomes 2H and 5H in both populations. One region on chromosome 5H was highly significantly associated with resistance to stripe rust (R 2 = 68% and 34% in TK and AF, respectively). The QTL on chromosome 2H accounted for 36% of the variation in TK and 10% of the variation in the AF population. These associations were consistent over both years. Further work will involve screening for additional markers in the target regions to identify polymorphism that can be used to select for multiple resistances in the absence of the pathogen.A R 0 2 2 3 1 M a p p i n g g e n e s f o r s t r i p e r e s i s t a n c e i n b a r l e y M . RESUMOAo lado dos projetos de seqüenciamento e das análises do cariótipo pelas técnicas de hibridização in situ, o desenvolvimento de mapas genéticos fundamentados em marcadores de DNA tem propiciado consideráveis avanços à genômica de plantas. Esta revisão aborda as premissas básicas utilizadas para o mapeamento genético e suas principais aplicações, especialmente para o melhoramento vegetal. Fundamentos teóricos sobre segregação, recombinação e ligação são considerados e relacionados à construção de mapas genéticos com marcas moleculares. Apresentam-se informações sobre tipos de marcadores, populações de mapeamento, cálculo da freqüência de recombinação, distorções da segregação, estabelecimento dos grupos de ligação e da ordenação dos marcadores. Discute-se, também, o uso de mapas de ligação em programas de seleção assistida por marcadores, na clonagem de genes e em estudos sobre sintenia. Palavras-chave: mapas de ligação, marcadores moleculares, QTL, mapeamento comparativo. ABSTRACT GENETIC MAPS IN PLANTSIn addition to genome projects and karyotype analysis by in situ hybridization techniques, a major advance in plant genome analysis came from the development of genetic maps based on molecular markers (Figure 1). This review clarifies the basic premises used for genetic mapping and its main applications, especially in plant breeding. The theories of segregation, recombination and linkage are considered and related to the construction of genetic maps based on molecular markers. Information about marker types, population mapping, calculation of the recombination rate, segregation distortion, linkage groups and genetic order determination is presented. Exploitation of linkage mapping for marker assisted selection, gene cloning and synteny comparisons is discussed.

Résumé : Après avoir fait une rapide revue des résultats disponibles à ce jour sur l'utilisation des marqueurs dans les expériences de rétro-croisement, nous nous intéresserons à l'efficacité de la SAM pour la prédiction des valeurs génétiques en nous appuyant notamment sur les résultats préliminaires d'une expérience de SAM en cours à la station de génétique végétale du Moulon. Summary :Marker-assisted selection (MAS) has received extensive attention in the past ten years. Two main strategies of MAS have been proposed: the use of markers to control the introgression of gene or QTL (quantitative trait locus), and the use of markers to predict breeding values in a population under selection. Theoretical and experimental results published on the interest of these two strategies are presented, focusing on the preliminary results of a MAS experiment performed on a maize population. From these results it appears that markers can be very efficient to quickly fix favorable alleles at target genes. Nevertheless, potential interactions between QTL (or genes) and environmental conditions or genetic background can reduce the efficiency of MAS. Further work is needed to optimize the use of markers in selection.
